【摘要】:药物成瘾已成为人们普遍关注的社会和医学难题。国外有学者认为,药物依赖是一种脑病,且与高血压、哮喘、Ⅱ型糖尿病三个公认的慢性病相比,在遗传性、个人责任、反复发作、治疗反应等方面有许多相似之处。因此药物依赖是一种需要医药治疗的慢性疾病。近来研究发现,药物成瘾是由于外源性的使用阿片类药物而抑制了体内神经元分泌内啡肽,因而一旦停药就可因体内的内啡肽下降而出现心理和生理的戒断反应。这已成为药物依赖的重要机制之一。随着细胞移植技术的发展,如果能将体外培养的具有稳定分泌β-内啡肽的细胞移植入药物成瘾者体内,植入的细胞就象一个“生物泵”源源不断的产生β-内啡肽,代偿由于药物依赖而产生的β-内啡肽分泌不足,使机体β-内啡肽达到正常水平,获得心理和生理的满足。这将成为治疗药物成瘾最理想的方法。本研究通过实验:1)建立大鼠下丘脑细胞分离和培养方法,观察体外培养的大鼠下丘脑细胞的生长、成熟、停滞的形态学特点和免疫细胞学鉴别;2)通过ELISA法,测定体外培养的大鼠下丘脑细胞分泌的β-内啡肽含量,了解下丘脑细胞分泌β-内啡肽的自身调节功能,进一步推断阿片类药物的成瘾机制;3)通过对体外培养的下丘脑细胞的冻存-复苏,了解其形态和分泌β-内啡肽功能的改变,探讨细胞移植治疗药物成瘾
[Abstract]:Drug addiction has become a common social and medical problem. Some foreign scholars believe that drug dependence is a brain disease, and compared with hypertension, asthma, type 2 diabetes in the three recognized chronic diseases, in the genetic nature. There are many similarities in personal responsibilities, recurring episodes, and therapeutic responses. Drug dependence is therefore a chronic disease that requires medical treatment. Recent research has found. Drug addiction is due to the exogenous use of opiates to inhibit the production of endorphins in neurons. Therefore, once the drug is stopped, psychological and physiological withdrawal can occur due to the decrease of endorphin in the body. This has become one of the important mechanisms of drug dependence. With the development of cell transplantation technology. If cultured cells with stable 尾 -endorphins are transplanted into drug addicts, the implanted cells are like a "biological pump" that keeps producing 尾 -endorphins. Compensatory 尾 -endorphin secretion due to drug dependence is insufficient, making the body 尾 -endorphin to the normal level. To obtain psychological and physiological satisfaction. This will be the best way to treat drug addiction. In this study, we established a method for the separation and culture of rat hypothalamic cells by experiment: 1. The growth, maturation and stagnation of rat hypothalamic cells in vitro were observed. 2) the content of 尾 -endorphin secreted by rat hypothalamic cells in vitro was measured by ELISA method to understand the self-regulation function of 尾 -endorphin secreted by hypothalamic cells. Further infer the mechanism of opioid addiction; 3) through cryopreservation and resuscitation of hypothalamic cells cultured in vitro, to understand the changes of morphology and 尾 -endorphin secretion, and to explore the treatment of drug addiction by cell transplantation.
