【摘要】:病毒性疾病的危害已威胁到人类生存,在2006年3月由国际医学科学院组织召开的第二届全球大会上,500名专家指出:不断变化的病毒使全球处于大流行的危险之中。WHO等单位联合发布的《发展中国家疾病控制优先项目》中指出:人类卫生面临的四大挑战,其中第三项为新发病毒性疾病的大流行。 随着分子生物学理论和技术在病毒学研究中的应用,尽管现代医学对病毒感染机制的认识有了飞越性的发展,但控制手段始终没有突破性进展,寻找有效的控制病毒性疾病的手段已成为21世纪人类的重要事件。 中医药防治病毒性疾病有数千年的经验和完整的理论体系,是中医药治病的优势领域。以其疗效确切、相对广谱、低毒的特点已被世人所接受,医学界期待从中医药中发现控制病毒性疾病的有效药物。近20年来,虽然大量的试验证实了中药治疗病毒性疾病的有效,尤其是SARS以后国内外从中药中寻找有效的抗病毒成分成为热点,但直至目前抗病毒中药的研制并没有突破性进展。其原因①缺乏充分体现中医药治疗病毒性疾病特点的研究思路;②缺乏充分体现中医药治疗病毒性疾病特点的研究手段。因此,在中医药治疗病毒性疾病理论的指导下,应用现代生物学技术和手段研究中药治疗病毒性疾病的机理,对中药走向世界具有重要的现实意义。 本研究在原有抗病毒药效试验和抗病毒作用机理研究的基础上,从病毒吸附环节的一些非特异性病理生理环节入手,以2~3种病毒株为样本,进一步探讨病毒吸附细胞的生物学机制和中药栀子提取物ZG和半枝莲总黄酮对此环节的干预作用,为寻找中药抗病毒作用的真正靶点奠定基础。本论文的研究内容为国家自然科学基金面上项目资助课题的部分内容。 一、栀子提取物ZG及半枝莲总黄酮的体外抗病毒作用研究 以Hep-2细胞株作为宿主细胞,以阿昔洛韦作为抗单纯疱疹病毒1型的阳性对照药,采用致细胞病变法(cytopathic effect CPE)来观察栀子提取物ZG和半枝莲总黄酮对4种病毒致细胞病变作用的影响,并按Reed-Muench方法计算最大无毒浓度(TC_0),50%细胞毒性浓度(TC_(50)),50%抑制浓度(IC_(50))和治疗指数(TI)。结果显示:栀子提取物
[Abstract]:The harm of viral diseases has threatened the survival of human beings at the second Global Congress organized by the International Academy of Medical Sciences in March 2006. 500 experts noted that the changing virus placed the world at risk of a pandemic. The World Health Organization (WHO), among others, reported in the priority for Disease Control in developing countries that:. Four major challenges to human health. The third is a pandemic of new viral diseases. With the application of molecular biological theory and technology in virology research, although the understanding of viral infection mechanism in modern medicine has developed by leaps and bounds, there has been no breakthrough in control methods. To find effective means to control viral diseases has become an important event in 21th century. Chinese medicine has thousands of years of experience in preventing and treating viral diseases and a complete theoretical system, which is the dominant field of TCM treatment. Because of its effective, relatively broad spectrum, low toxicity has been accepted by the world. The medical profession is looking forward to the discovery of effective drugs for the control of viral diseases from Chinese medicine. In the past 20 years, although a large number of experiments have proved the effectiveness of Chinese medicine in the treatment of viral diseases. Especially after SARS, it has become a hot spot to look for effective antiviral components from traditional Chinese medicine at home and abroad. However, until now, there has been no breakthrough in the development of antiviral Chinese medicine. The reasons are: 1 lack of research ideas to fully reflect the characteristics of traditional Chinese medicine in the treatment of viral diseases; 2 lack of research means to fully reflect the characteristics of traditional Chinese medicine in the treatment of viral diseases. Therefore, under the guidance of the theory of traditional Chinese medicine treatment of viral diseases. The application of modern biological techniques and means to study the mechanism of traditional Chinese medicine in treating viral diseases is of great practical significance to the development of traditional Chinese medicine in the world. On the basis of the original antiviral effect test and the mechanism of antiviral action, this study began with some non-specific pathophysiological links of virus adsorption, and took three virus strains as samples. To further explore the biological mechanism of virus adsorption cells and the effect of ZG and total flavonoids of Gardenia jasminoides on this link. In order to find the real target of antiviral effect of traditional Chinese medicine, the research content of this paper is part of the project supported by the National Natural Science Foundation of China. Antiviral effects of Gardenia jasminoides extract ZG and total flavonoids of Scutellaria barbata in vitro Hep-2 cell line was used as host cell and acyclovir as positive control drug against herpes simplex virus type 1. Cytopathic effect was used to observe the effects of Gardenia jasminoides extract ZG and total flavonoids of Scutellaria barbata on cytopathic effects of four viruses. The maximum nontoxic concentration (TC0%) was calculated by Reed-Muench method. 50% inhibitory concentration (ICS) and treatment index (TI). The results showed that the extract of Gardenia jasminoides was extracted from Gardenia jasminoides.
