【摘要】:实验目的:建立大鼠湿困脾胃证动物模型及其生物学特征。 实验方法:采用增加湿度、饮食调控结合睡眠控制的方法建立大鼠湿困脾胃证动物模型,选择经典方药平胃散作为反证治疗药物;用定量、半定量方法观察大鼠湿困脾胃证动物模型的宏观体征;用半固体营养糊法测定大鼠的胃和小肠运动功能变化;用离子选择电极法测定血清电解质;用GOD-PAP法测定血糖(Glu);用双缩脲法测定总蛋白(TP);用GPO-PAP法测定甘油三脂(TG);用CHOD-PAP法测定总胆固醇(TC);用ELISA法测定血清胃泌素(Gas)、免疫球蛋白-G(IgG)、胰岛素(Insulin)、白细胞介素-6(IL-6);用放免法检测血清胃动素(MTL);用比色法测定肝细胞Na~+-K~+-ATP酶活性;用HE染色法观察脾、胸腺及胃肠组织形态学变化。 实验结果:1 大鼠湿困脾胃证动物模型及其生物学特征的建立①宏观体征:模型形成时间较长;难以速愈,模型体征短期内不易恢复;模型大鼠生长状态不佳,皮毛湿浊色泽晦暗发黄,消瘦,体重减轻,体长、尾长、尾根直径增长缓慢;食欲不振,饮食量减少;小便黄量少,大便溏湿重增加;腹胀,腹围指数增加;精神萎靡,阴囊松弛下垂,嗜卧懒动,自发活动次数减少;口粘腻,口腔PH值微升;体温微升或不变;与空白对照组比较有改变趋势或有显著性差异(P<0.05或P<0.01)。②胃肠功能:模型动物胃肠胀气明显,胃排空和小肠推动能力下降,胃肠吸收功能异常,胃肠激素水平紊乱,胃肠组织发生病理改变,与空白对照组比较有改变趋势或有显著性差异(P<0.05或P<0.01)。③物质能量代谢:模型大鼠脏器系数增加,血清电解质水平紊乱,,糖、蛋白、脂类含量异常,胰岛素水平下降,Na~+-K~+-ATP酶活性降低,与空白对照组比较有改变趋势或有显著性差异(P<0.05或P<0.01)。④免疫功能:模型脾脏及胸腺脏器系数降低,胸腺皮质厚度及脾脏中央动脉淋巴鞘直径、面积变小,血清IL-6升高,IgG含量明显减少,与空白对照组比较有改变趋势或有显著性差异(P<0.05或P<0.01)。2 反证药物平胃散对该模型的治疗作用:经平胃散治疗后,治疗组大鼠生长良好,皮毛浓密而有光泽,在宏观体征、胃肠功能、物质能量代谢、免疫功能四方面的多数指标均有所改善或恢复正常,与模型组、自然恢复组比较有改变趋势或有显著性差异(P<0.05或P<0.01)。 实验结论:本实验制备的大鼠湿困脾胃证动物模型,从宏观体征、胃肠功能、物质能量代谢、免疫功能四个方面具有显著的生物学特征,经典方药平胃散对该模型良好的治疗作用也说明了模型的成功性。因此我们认为采用增加湿度、饮食调控结合睡眠控制的方法制备的大鼠湿困脾胃证动物模型是一个比较符合中医理论的动物模型。
[Abstract]:Objective: to establish an animal model of dampness of spleen and stomach and its biological characteristics. Methods: the animal model of spleen and stomach dampness was established by increasing humidity, diet regulation and sleep control. The macroscopical signs of rat model of dampness in spleen and stomach were observed by quantitative and semi-quantitative methods. The changes of gastric and intestinal motor function in rats were measured by semisolid nutritional paste method. Serum electrolytes were determined by ion selective electrode method. Blood glucose was determined by GOD-PAP method. The total protein TPX was determined by biuret method. GPO-PAP method was used for the determination of triglyceride. Total cholesterol (TC) was determined by CHOD-PAP method. The levels of serum gastrin, immunoglobulin G, insulin, interleukin 6 and interleukin 6 were determined by ELISA method. The serum motilin motilin (MTL) was detected by radioimmunoassay. The activity of Na ~ -K ~ -ATPase in hepatocytes was measured by colorimetry. The morphological changes of spleen, thymus and gastrointestinal tissues were observed by HE staining. Results the animal model of dampness of spleen and stomach and its biological characteristics were established. 1 macroscopic signs: the formation time of the model was longer; It is difficult to recover quickly and the physical signs of the model are not easy to recover in a short time. The growth state of the model rats was not good, the fur was dark and yellow, thin, weight loss, body length, tail length, tail root diameter growth slow; Loss of appetite and reduced diet; Urination Huang Liang little, loose stools wet weight increase; Abdominal distension, abdominal circumference index increased; The spirit is dismal, scrotum flabby and drooping, lying lazy and moving, and the number of spontaneous activities decreases; The mouth is sticky and greasy, and the PH value of the mouth increases slightly; Slight rise or no change in body temperature; Compared with the blank control group, there were significant differences in gastrointestinal function (P < 0.05 or P < 0.01). Abnormal gastrointestinal absorption function, gastrointestinal hormone level disorder, gastrointestinal tissue pathological changes. Compared with the blank control group, there were significant differences in substance and energy metabolism (P < 0.05 or P < 0.01). The lipid content was abnormal, the insulin level was decreased and the Na ~ -K ~ -ATPase activity was decreased. Compared with the blank control group, the immune function of the model was significantly different (P < 0.05 or P < 0.01). The spleen and thymus organ coefficients of the model were lower than that of the blank control group (P < 0.05 or P < 0.01). The thickness of thymic cortex and the diameter and area of lymphatic sheath of the central splenic artery became smaller, and the content of serum IL-6 increased significantly. Compared with the blank control group, there were significant differences (P < 0.05 or P < 0.01). 2 the therapeutic effect of Pingwei Powder on the model: after treatment with Pingwei Powder. The treatment group rats grew well, fur thick and shiny, in the macro signs, gastrointestinal function, substance and energy metabolism, immune function of the four indicators are improved or returned to normal, with the model group. In natural recovery group, there was a change trend or significant difference (P < 0.05 or P < 0.01). Conclusion: the animal model of dampness of spleen and stomach in this experiment has significant biological characteristics in four aspects: macroscopic signs, gastrointestinal function, substance and energy metabolism, and immune function. The good therapeutic effect of classical prescription Pingwei Powder on the model also shows the success of the model. The animal model of dampness of spleen and stomach was made by the method of diet regulation and sleep control.
