复合因素塑造劳倦过度脾虚证动物模型的实验研究

【摘要】:自1980年北京师范大学生物系首次用大黄复制脾虚证动物模型以来,已有十多种动物模型相继建立。动物模型的建立,使关于脾虚证较深入的实验研究成为可能。目前研究者已对各种脾虚证动物在消化吸收、免疫、神经、内分泌等方面的改变进行了探讨。但是,实验研究中常用的脾虚证动物模型与临床脾虚证还存在一定的差距。传统中医认为“劳”作为病因包括劳力过度、劳神过度和房劳过度三方面,而现有的“劳倦过度”脾虚证动物模型,主要采用力竭运动造成动物身体疲劳,对动物精神的影响不明显。所谓“劳倦过度”造模方法仅模拟了劳力过度,没有模拟劳神过度这一因素,与临床脾虚证有一定差距。另外,多数脾虚证造模方法为单因素造模,仅有少数几种复合因素脾虚证模型。因此,对现有“劳倦过度”脾虚证单因素动物模型进行改善,建立一个比较接近临床脾虚证的动物模型已成为脾虚证本质研究的迫切需求。 为了更好地模拟劳力过度和劳神过度两方面的致病原因,本实验在以往力竭游泳单因素塑造脾虚证动物模型的基础上剥夺动物的部分睡眠时间,使动物不仅劳力而且劳神。从而建立了一种新的脾虚证动物模型——改良型劳倦过度小鼠脾虚证模型。 实验主要观察了动物的一般体征和消化吸收功能的变化,测定的指标有:D—木糖排泄率、血清淀粉酶活性、胃排空速率、肠推进速率、离体小肠张力、胃肠粘膜形态、血清胃泌素含量、脾脏指数和胸腺指数。通过与大黄泻下法、力竭游泳法塑造的脾虚证动物模型比较、药物反证等几方面来评估和验证此方法的成立,并对其影响动物消化系统功能的机制作了初步探讨。 实验结果显示,改良型劳倦过度模型小鼠造模后出现脾虚证的典型外观表现,且客观指标改变与文献报导相符,与临床脾虚证患者变化趋势一致。在所选的指标中,改良型劳倦过度组小鼠的变化均与临床脾虚证患者的变化趋势一致,且多数指标的变化较大黄组(大黄泻下法塑造脾虚证动物模型)和力竭游泳组(力竭游泳法塑造脾虚证动物模型)明显,而大黄组和力竭游泳组只有部分指标的变化与临床患者一致,因此,改良型劳倦过度法塑造脾虚证动物模型优于大黄泻下法和力竭游泳法。运用四君子汤治疗进行反证,小鼠一般体征、各客观指标较自然恢复组明显改善,说明改良型劳倦过度法塑造的脾虚证模型符合脾虚证动物模
[Abstract]:Since 1980, when the animal model of spleen deficiency syndrome was first replicated with rhubarb in the Department of Biology of Beijing normal University, more than ten animal models have been established one after another. At present, researchers have discussed the changes in digestion, absorption, immunity, nerve, endocrine and other aspects of various spleen deficiency syndrome animals. There is still a certain gap between the animal model of spleen deficiency syndrome and clinical spleen deficiency syndrome. Traditional Chinese medicine believes that "labor" as the etiology includes three aspects: excessive labor, excessive mental exhaustion and excessive room labor. However, the existing animal model of "overfatigue" of spleen deficiency syndrome mainly uses exhausted exercise to cause animal body fatigue, which has no obvious effect on animal spirit. The so-called "overfatigue" modeling method only simulates overwork. There is a certain gap between the model of spleen deficiency syndrome and clinical spleen deficiency syndrome. In addition, most of the methods of making model of spleen deficiency syndrome are single factor model, only a few kinds of multiple factors spleen deficiency syndrome model. It is an urgent need to study the essence of spleen deficiency syndrome by improving the existing single factor animal model of spleen deficiency syndrome and establishing an animal model which is close to clinical spleen deficiency syndrome. In order to better simulate the causes of overwork and overwork, this experiment deprived the animals of part of their sleep time on the basis of the previous exhaustive swimming single factor model of spleen deficiency syndrome. A new animal model of spleen deficiency syndrome, modified model of spleen deficiency in overworked mice, was established. The changes of general signs and digestive and absorption functions of animals were observed. The indexes were: excretion rate of 1: D- xylose, serum amylase activity, gastric emptying rate and intestinal propulsion rate. Small intestinal tension, gastrointestinal mucosal morphology, serum gastrin content, spleen index and thymus index were compared with the animal model of spleen deficiency by using the method of rhubarb diarrhea and exhaustive swimming. The establishment of this method was evaluated and verified in several aspects, and the mechanism of its influence on the digestive system of animals was preliminarily discussed. The results showed that the typical appearance of spleen deficiency syndrome appeared in the modified model mice, and the change of objective indexes was consistent with the literature reports. In the selected indexes, the changes of the modified overfatigue group were consistent with the changes of the clinical spleen deficiency syndrome patients. The changes of most indexes were significant in yellow group (rhubarb diarrhea model) and exhaustive swimming group (exhaustive swimming method for spleen deficiency animal model). The rhubarb group and exhaustive swimming group only part of the changes in the same as the clinical patients, so. The modified model of spleen deficiency syndrome was better than that of rhubarb diarrhea and exhaustive swimming. The general signs and objective indexes of mice were obviously improved compared with the natural recovery group. It shows that the model of spleen deficiency is consistent with the animal model of spleen deficiency.
