腺苷A1受体对新生鼠离体延髓脑片呼吸节律性放电及呼吸神经元电活动的调制

【摘要】:目的 1) 应用新生大鼠离体延髓脑片标本,记录与之相连的舌下神经呼吸节律性放电(respiratory rhythmical discharge activity,RRDA),观察腺苷A1受体的特异性拮抗剂8-环戊-1,3-二丙基黄嘌呤(8-cyclopentyl-1,3-dipropylxa nthine,DPCPX)和特异性激动剂 R-苯异丙基-腺苷(R-phenylisoprpyl-adenosine,R-PIA)对RRDA的影响,从而探讨腺苷A_1受体在节律性呼吸的产生和调节中的作用;2) 在新生大鼠离体延髓脑片标本上,同步记录舌下神经根和面神经后核内侧区(the medial area of retrofacialis nucleus,mNRF)的双相呼气神经元/吸气神经元的放电活动,观察腺苷A1受体的特异性拮抗剂和激动剂对呼吸神经元放电活动的影响,进一步探讨腺苷A1受体对节律性呼吸的调制。 方法 选用新生SD大鼠(0~2d),雌雄不拘。1) 参照并改良Suzue的方法制作离体延髓脑片标本,主要包含面神经后核内侧区,前包钦格氏复合体(Pre-B(?)tzinger Complex)、腹侧呼吸组以及背侧呼吸组的一部分,保留舌下神经根。用玻璃吸附电极记录舌下神经放电作为呼吸活动的指标,通过灌流液给药,观察不同药物对神经根放电活动的影响;2) 在面神经后核内侧区(mNRF)同步记录双相呼气神经元/吸气神经元的放电活动,灌流给药,观察其对呼吸神经元放电的影响。 结果 1.记录舌下神经根放电的实验部分:1) DPCPX和R-PIA对RRDA的影响:与给药前对照相比,应用2μmol/L DPCPX后,呼吸周期(RC)显著性降低,呼气时程(TE)显著性缩短,RC缩短27.07%(P0.05),TE减少28.38%(P0.05),而吸气时程(TI)和放电的积分幅度(IA)较对照无明显变化,第15min的结果较第10min的变化不大。改用10μmol/L R-PIA灌流后15min
[Abstract]:Objective 1) to apply in vitro medulla oblongata of newborn rats. Respiratory rhythmic discharges associated with the hypoglossal nerve (rhythmical discharge activity) were recorded. To observe the specific antagonist of adenosine A1 receptor, 8-cyclopentyl-1-dipropylxa nthine. DPCPX and R-phenylisopr-adenosine R-PIA-a specific agonist on RRDA. Therefore, the role of adenosine A-1 receptor in the production and regulation of rhythmic respiration was discussed. 2) on the medulla oblongata of newborn rats. The medial area of retrofacialis nucleus were recorded simultaneously in the root of the hypoglossal nerve and the medial area of the posterior nucleus of the facial nerve. The effects of specific antagonists and agonists of adenosine A1 receptor on the activity of respiratory neurons were observed. To further investigate the modulation of adenosine A1 receptor on rhythmic respiration. Methods the medulla oblongata slices of neonatal SD rats were made by using the method of modified Suzue, including the medial area of the posterior nucleus of the facial nerve, according to the method of using male and female Sprague-Dawley rats for 2 days. Pre-Bu? The hypoglossal nerve root was preserved in the ventral breathing group and the dorsal breathing group. The hypoglossal nerve discharge was recorded with the glass adsorption electrode as the index of respiratory activity. The effects of different drugs on the activity of nerve root discharge were observed. 2) the discharges of biphasic expiratory / inspiratory neurons were recorded synchronously in the medial region of the posterior nucleus of the facial nerve, and the effects of MNRFs on the discharges of respiratory neurons were observed. Results 1. The effects of DPCPX and R-PIA on RRDA were recorded in the experimental part of hypoglossal nerve root discharges: compared with those before administration. After 2 渭 mol/L DPCPX, the respiratory cycle was significantly decreased, the expiratory duration was significantly shortened and RC was shortened by 27.07% (P 0.05). Te decreased 28.38% (P 0.05), but the inspiratory time interval (tie) and the integral amplitude of discharge (IAA) showed no significant change compared with the control group.
