胸腺内注射异基因骨髓细胞诱导特异性免疫耐受的实验研究

【摘要】:目的: 探索通过胸腺内注射异基因骨髓细胞加腹腔注射环磷酰胺,以小鼠皮肤移植为模型,诱导移植免疫耐受,为免疫耐受提供理论和实验依据。 方法: 提取供体C57BL/6(B6)小鼠(H-2~b)骨髓细胞,注入受体BALB/C(H-2~d)小鼠胸腺内,48h后腹腔注射200mg/kg的环磷酰胺,2w后移植供体C57BL/6(B6)小鼠(H-2b)和无关供体C3H(H-2~k)小鼠皮肤。共设空白对照组、CP对照组、BM对照组、实验组、第三方供体组5个组。观察皮肤存活时间;皮肤移植2w后对受体BALB/C(H-2~d)小鼠作单向混合淋巴细胞反应(mixed lymphocyte reaction,MLR)、脾细胞增值反应(Spleen cells proliferation reaction)及迟发型超敏反应(delayed type hypersensitivity,DTH)耐受状态的检测。 结果: 耐受BALB/C小鼠获得了对供体B6小鼠的特异性皮肤移植耐受,实验组移植皮肤平均存活时间(MST)为22.00±2.51天,对照组MST为8.75±1.04天,耐受的BALB/C小鼠对第三供体C3H小鼠的移植皮肤仍正常排斥,MST为10.25±1.91天。单向混合淋巴细胞反应和迟发型超敏反应证明耐受的BALB/C小鼠对B6小鼠的脾细胞的反应性特异性降低,而仍保持对无关第三者供体C3H小鼠的免疫反应。耐受的BALB/C小鼠脾细胞对丝裂原(ConA)的增殖反应在正常范围。 结论: 使用胸腺内注射供体异基因骨髓细胞,结合腹腔应用环磷酰胺的
[Abstract]:Objective: to explore the induction of allogeneic immune tolerance by intrathymic injection of allogeneic bone marrow cells and intraperitoneal injection of cyclophosphamide in mice, and to provide theoretical and experimental evidence for immune tolerance. Methods: bone marrow cells were extracted from donor C57BL / 6 B6) mice and injected into the thymus of the recipient BALB / CnH-2B) mice. After 48 hours, 200 mg / kg cyclophosphamide was injected intraperitoneally. After 2 weeks, the donor C57BL / 6 B6 mice were transplanted into the skin of C57BL / 6B6) and the unrelated donor C3H2H 2) mice. The control group was divided into two groups: the CP control group and the BM control group, the experimental group were divided into two groups: the control group and the control group.
