间充质干细胞抑制T淋巴细胞增殖减轻移植物抗宿主病机制研究

【摘要】:背景与目的: 间充质干细胞(Mesenchymal stem cells,MSCs)是骨髓非造血细胞中成体干细胞中的一种,具有很强的自我更新能力和多向分化潜能。在体外不同分化条件的诱导下,可以形成成骨细胞、软骨细胞或脂肪细胞等,并且克隆后的细胞具有类似分化潜能,充分证明了间充质干细胞是多能分化细胞。除在骨髓中发现间充质干细胞外,在骨骼肌、心肌、真皮,以及脑和外周血中,也发现有类似骨髓间充质干细胞(BM-MSCs)生物学特性和表面标记的间充质干细胞。间充质干细胞表型不均一,,分别具有间充质细胞、上皮细胞和内皮细胞的特点。对人体外扩增的BM—MSCs的研究结果显示一致表达SH2、SH3、CD29、CD44、CD77、CD90、CD106、CD120a、CD124等抗原,不表达造血干细胞(HSC)的标志性抗原CD14、CD34、CD45。但是目前还没有特异的表面标记来证实间充质干细胞。动物实验和临床研究均发现,异基因造血干细胞移植时共同输入供体间充质干细胞可以降低移植失败率并减轻移植物抗宿主病(GVHD)。具体机制尚不明确,大多数学者认为间充质干细胞可能通过抑制T淋巴细胞增殖来减轻移植后GVHD,而这种抑制作用是通过分泌可溶性细胞因子实现的,最受关注的细胞因子是转化生长因子-β_1(TGF-β_1)和肝细胞生长因子(HGF)。 本实验通过体外培养间充质干细胞(Mesenchymal stem cells,MSCs)探索其鉴定方法,观察间充质干细胞的生物学特性及TGF-β_1和HGF的表达,通过与T淋巴细胞的混合培养研究其抑制T淋巴细胞增殖,减轻GVHD的机制。 材料与方法:

Animal experiments and clinical studies have found that, Allogeneic hematopoietic stem cell transplantation combined with donor mesenchymal stem cells can reduce graft failure rate and alleviate graft versus host disease (GV HDD). The mechanism is not clear. Most researchers believe that mesenchymal stem cells may reduce GVHD after transplantation by inhibiting T lymphocyte proliferation, which is achieved by secreting soluble cytokines. The most concerned cytokines were transforming growth factor- 尾 (TGF- 尾) and hepatocyte growth factor (HGF- 尾). In this experiment, mesenchymal stem cells were cultured in vitro to explore the identification of mesenchymal stem cells. The biological characteristics of mesenchymal stem cells and the expression of TGF- 尾 1 and HGF were observed. The proliferation of T lymphocytes was inhibited by co-culture with T lymphocytes.
