CK-794致小鼠毛发生长发育异常动物模型的建立及其基因表达谱的分析

【摘要】:本研究从给药剂量、给药时间、性状有无遗传性几个方面进行研究,建立了两种品系小鼠毛发生长发育异常的动物模型。并从仔鼠毛发的生长及分布情况、毛发类型、毛发长度、毛发直径及毛发密度几个方面对各组仔鼠的毛发性状进行了描述。10mg/kg组仔鼠在给药期肉眼未见毛发生长。在此基础上,用小鼠全基因组芯片对接触药物组仔鼠及对照组仔鼠皮肤毛囊组织的基因表达谱进行了分析,发现大部分编码构成毛发的蛋白质的相应基因(Krt及Krtap)在给药组仔鼠的皮肤毛囊组织中的表达明显下调,而且参与Krt及Krtap表达调控的基因也发生了明显变化。在此基础上对CK-794诱发的毛发生长发育异常可能的机制进行初步分析。
[Abstract]:In this study, animal models of abnormal hair growth and development in two strains of mice were established from the aspects of dosage, time and heredity of administration, and the growth and distribution of hair and hair types in two strains of mice were also studied. Hair length, hair diameter and hair density were used to describe the hair characteristics of rats in each group. 10 mg / kg group had no hair growth in the naked eye during the administration period. The gene expression profiles of the skin hair follicles of the mice exposed to the drug and the control group were analyzed by using the whole genome microarray of mice. It was found that the expression of most of the corresponding genes, Krt and Krtapin, which encodes the proteins that constitute hair, was significantly down-regulated in the skin hair follicles of the rats in the administration group. Moreover, the genes involved in the regulation of Krt and Krtap expression also changed obviously. The possible mechanism of hair growth and development abnormality induced by CK-794 was analyzed.
