冠脉结扎法建立大鼠急性心肌梗死模型方法的探讨

【摘要】:目的建立一种方便、省时、死亡率低且稳定的急性心肌梗死模型制作方法。方法雄性成年Sprague·Dawley大鼠麻醉后在呼吸机支持下,于相应第3、4胁间隙处开胸,挤压两侧胸壁,将心脏挤出于胸腔外,通过结扎左冠状动脉前降支,建立心肌梗死模型。通过观察术后大鼠心电图的变化,以及分别利用Evan blue染色及病理组织学方法于术后3 h及1周,确定心肌缺血及梗死范围,同时观察术中、术后24 h及1周大鼠的存活率。结果术后大鼠Ⅰ、Ⅱ导联心电图ST段明显抬高。术后3 h大鼠心脏Evan blue染色显示心肌缺血区恒定。术后1周处死取出大鼠心脏,可见心脏前壁、侧壁及心尖处形成境界清晰的心肌梗死区,梗死区颜色明显浅于周围的心肌,呈苍白色,失去正常光泽。术后1周大鼠心肌组织HE染色,可见梗死区伴有大量炎性细胞浸润。造模成功率为83%,术后24 h存活率为77%,术后1周大鼠总存活率为72%。结论此模型操作简单、稳定,且术后动物存活率高,是研究心肌梗死较理想的模型。
[Abstract]:Objective to establish a convenient, time-saving, low mortality and stable model of acute myocardial infarction (AMI). Methods male adult Sprague 路Dawley rats were anesthetized and treated with ventilator. The myocardial infarction model was established by ligating the anterior descending branch of the left coronary artery by squeezing the heart out of the chest. The changes of electrocardiogram (ECG) of the rats were observed after operation, and Evan blue staining and histopathology were used at 3 h and 1 week after operation, respectively.
