用hTSHR活化的脾细胞免疫小鼠建立甲状腺相关性眼病模型的研究

【摘要】:目的 用 hTSHR 活化的脾细胞免疫同源 BALB/c 小鼠建立甲状腺相关性眼病(TAO)动物模型。 方法 将 6-8 周雌性 BALB/c 小鼠预处理后随机分为两组(a 组和b 组),分别用重组质粒 pcDNA3.1/hTSHR 和空质粒 pcDNA3.1 免疫两组小鼠,并在免疫后的第 3 周、第 6 周加强免疫两次。18 周后分别处死两组小鼠,取眼眶组织做病理学观察,并机械分离脾细胞。将 6-8 周雌性 BALB/c 小鼠随机分为两组(c 组和 d 组),分别接受 a 组和 b 组小鼠的脾细胞免疫。4 周后处死小鼠,取眼眶组织做病理学观察,用放射免疫法测定血清 TT4 水平、免疫放射法测定血清 TSH 水平,ELISA 法检测血清 TRAb。 结果 a 组(重组质粒免疫组):约 25%的小鼠眼眶组织出现粘液变性、水肿,但无明显纤维组织及脂肪组织增生,肌纤维无变性断裂。c 组(活化脾细胞免疫组):约有 50%的小鼠眼眶组织出现明显粘液变性、水肿,局部纤维组织和脂肪组织增生,肌纤维透明变性断裂;电镜下可见滑面内质网扩张,肌原纤维排列紊乱。b 组(空质粒免疫组)和 d 组(未活化脾细胞免疫组):小鼠的眼眶组织均无形态学改变。脾细胞免疫后 4 周,血清 TT4:c 组(7.130±1.017 ug/dl)与 d 组(6.431±0.573 ug/dl)相比,TT4 水平显著升高(p=0.036)。血清 TSH:c组(0.070±0.032 uIU/ml)与 d 组(0.098±0.020 uIU/ml)相比,TSH 水平显著降低(p=0.020)。其中,2 只小鼠的血清 TT4 高于参考范围上限,血清 TSH 低于参考范围下限,符合甲状腺功能亢进的血清学
[Abstract]:Objective to establish an animal model of thyroid associated ophthalmopathy (TAO) in BALB/c mice immunized with spleen cells activated by hTSHR. Methods female BALB/c mice of 6-8 weeks were randomly divided into two groups: group A and group b. The mice were immunized with recombinant plasmid pcDNA3.1/hTSHR and empty plasmid pcDNA3.1 at the third week after immunization. After 6 weeks, the mice were killed and the orbital tissues were taken for pathological observation. Six to eight weeks old female BALB/c mice were randomly divided into two groups: group C and group d. The mice of group a and group b were immunized with spleen cells for 4 weeks, and then the mice were killed, and the orbital tissues were taken for pathological observation. Serum TT4 levels were measured by radioimmunoassay and serum TSH levels by immunoradiometric assay. Results in group a (recombinant plasmid immunized group), about 25% mice had mucus degeneration and edema, but there was no obvious proliferation of fibrous tissue and adipose tissue. In group c (activated splenocyte immunity group), about 50% of mouse orbital tissues showed obvious mucus degeneration, edema, proliferation of local fibrous tissue and adipose tissue, and hyaline degeneration of muscle fiber. Electron microscope showed smooth endoplasmic reticulum dilatation, myofibril arrangement disorder. Group b (empty plasmid immunized group) and group d (unactivated splenocyte immunization group): no morphological changes were found in the orbital tissues of mice. The spleen cells were immunized 4 weeks after immunization. Compared with d group (6.431 卤0.573 ugr / dl), the level of TT4 in serum TT4:c group was significantly higher than that in group d (0.070 卤0.032 渭 mol 路dll), and the level of TT4 in serum TSH:c group was significantly lower than that in group d (0.098 卤0.020 uIU-dll). The serum TT4 of 2 mice was higher than the upper limit of reference range. Serum TSH was lower than the lower limit of reference range, which was in accordance with the serology of hyperthyroidism.
