吉林省汉坦病毒的分离及进化分析

【摘要】:汉坦病毒(Hantavirus,HV)属于布尼亚病毒科(Bunyyaviridae)汉坦病毒属(Hantavirus),是一有包膜的分节段的负链RNA病毒。HV基因组由大(Large,L)、中(Medium,M)、小(Small,S)三个片段组成。L片断长6530-6550bp,编码病毒特异的依赖RNA的RNA聚合酶;M片断大小3613-3707bp,编码G蛋白前体(GPC);S片断由1696-2083bp组成,编码病毒的核衣壳蛋白(NP)。 汉坦病毒一个主要特征是每一型的HV都有一个独特的啮齿动物宿主(除TPM株以外)。汉坦病毒可在其宿主中长期存在而不表现症状,并与宿主共进化。人主要通过接触带毒宿主动物及其分泌物和排泄物所形成的气溶胶而感染。人感染HV后可引起两种死亡率很高的疾病:肾综合征出血热(Hemorrhagic Fever with Renal Syndrome,HFRS)和汉坦病毒肺综合征(Hantavirus Pulmony Syndrome,HPS)。 目前,已有23种汉坦病毒的血清型/基因型被确定。我国流行的主要是Ⅰ(HTN)型和Ⅱ(SEO)型汉坦病毒。最近有报导在我国发现普马拉(PW)型汉坦病毒。 本研究共分为两个部分: 1、用直接免疫荧光法检测从吉林省不同疫区采集的褐家鼠鼠肺标本,共得到55份阳性标本,阳性率为9.35%;将其中4个阳性标本(Jilin1050、Jilin1055、Jilin1056和Jilin1060)作RT-PCR检测,结果为SEO(Ⅱ)型病
