B7-H4基因修饰的胰岛细胞对小鼠同种异体胰岛移植排斥反应的影响

【摘要】: 本课题利用携带免疫负调控分子B7-H4的重组质粒转染自BABL/c小鼠分离得到的胰岛细胞,并采用肾包膜下注射的方法将其移植给诱发Ⅰ型糖尿病的C57BL/6小鼠模型,观察移植物的功能状态和存活时间,探索避免同种异体胰岛移植排斥反应的有效方法。在体外将基因修饰的胰岛细胞与T淋巴细胞混合培养,观察其对T细胞增殖和细胞因子分泌的影响。结果显示:该法能明显改善受体鼠糖尿病症状,有效控制血糖水平,显著延长同种异体移植胰岛的存活期;B7-H4基因修饰的胰岛细胞在体外刺激T淋巴细胞增殖的能力显著下降,并可明显抑制IL-2、IFN-γ的分泌。为Ⅰ型糖尿病的胰岛移植治疗研究提供了新的途径,为进一步的免疫负向调控分子的理论和应用研究提供了新的依据和方法。
[Abstract]:In this study, islet cells isolated from BABL/c mice were transfected with recombinant plasmid carrying B7-H4, and transplanted into C57BL/6 mice model of type 1 diabetes mellitus by subcapsular injection.To observe the functional state and survival time of grafts and to explore an effective method to avoid allograft rejection.The effects of gene modified islet cells and T lymphocytes on T cell proliferation and cytokine secretion were observed in vitro.The results showed that this method could significantly improve the symptoms of diabetes in recipient mice, effectively control the level of blood glucose, and prolong the survival period of islet allografts significantly. The ability of stimulating T lymphocyte proliferation of islet cells modified with B7-H4 gene was significantly decreased in vitro.It also inhibited the secretion of IFN- 纬.It provides a new approach for the study of islet transplantation in type I diabetes mellitus, and provides a new basis and method for the further study of the theory and application of immuno-negative regulation molecules.
