线虫磷脂酶D的生物学功能研究

【摘要】:磷脂酶D(Phospholipase, PLD)是细胞信号转导网络中的重要成员之一,与许多人类疾病都有密切的关系,是一个潜在的药物治疗靶标。但到目前为止,其生物学功能和作用机制尚未研究清楚,由于人类的细胞信号转导系统非常复杂,所以本研究选择了相对简单的模式生物——线虫,来研究PLD的生物学功能,并对其作用机制进行初步的探讨。线虫作为一种真核多细胞生物,其基因组序列的测定早在1998年就已经完成,经过多年研究发现,线虫存在着许多与高等生物类似的细胞信号转导通路,并且与其它多种生物模型相比,线虫还具有结构简单、饲养方便、生命周期短以及便于观测等许多优点。 在本研究中,我们以PLD-1基因敲除的线虫种系RB1040作为研究对象,通过比较其与野生型线虫N2在相同培养条件下表型(产卵数量、寿命、运动、吞咽、排便、化学趋势和食物趋势等)上的差异来推测PLD的生物学功能。同时还利用连接有绿色荧光蛋白(GFP)的PLD-1敲除型线虫来研究PLD-1的表达部位,初步探讨了PLD-1在线虫中的作用机制,为研究人PLD的生物学功能提供了理论依据。
[Abstract]:Phospholipase phospholipase (PLD) is one of the important members of cell signal transduction network and has close relationship with many human diseases and is a potential target for drug therapy.But up to now, its biological function and mechanism have not been studied. Because of the complexity of human cell signal transduction system, we chose a relatively simple model organism, nematode, to study the biological function of PLD.The mechanism of its action was discussed.As a kind of eukaryotic multicellular organism, the genome sequence of nematode has been completed since 1998. After many years of research, it has been found that nematodes have many cell signal transduction pathways similar to those of higher organisms.Compared with other biological models, nematode has many advantages, such as simple structure, convenient feeding, short life cycle and easy observation.In this study, we compared the phenotypes (egg number, longevity, movement, swallowing, defecation) of PLD-1 knockout nematode strain RB1040 under the same culture conditions as wild-type nematode N2.The differences in chemical and food trends are used to speculate on the biological function of PLD.At the same time, the PLD-1 knockout nematodes with green fluorescent protein (GFP) were used to study the expression of PLD-1, and the mechanism of PLD-1 online was discussed, which provided a theoretical basis for the study of the biological function of human PLD.
