新生期暴露于炔雌醇对雄性大鼠生殖系统的生长发育及功能的影响

【摘要】:雌激素主要通过与雌激素受体结合发挥生物学作用。在雄性动物的生殖系统中有大量的雌激素产生,适当浓度的雌激素对雄性生殖的生长发育和维持正常功能有非常重要的作用,雌激素缺乏或过高浓度的暴露都会引起对雄性动物生殖系统生长发育和功能的损害,但目前对雌激素在生殖系统中的作用机制仍然了解不多。由于环境中的雌激素和有雌激素活性的化学物质不断增多,使人们迫切的想知道雌激素对雄性动物生殖系统的作用位点和作用机制以及雄性动物和人暴露于生活中的环境雌激素或雌激素样物质是否会导致其生殖结构和正常功能受到不利的影响。本实验目的是为了探索在新生期暴露于雌激素,雄性大鼠生殖系统生长发育和功能受到的不利影响及其作用机制。方法:在出生第2天至12天期间,新生期雄性大鼠分组分别隔日给不同剂量:10μg/2d、500ng/2d和25ng/2d炔雌醇处理(处理组),对照组大鼠给予玉米油。饲养至成年后,处死,取材检测各组成年大鼠睾丸和附睾的一般形态结构、精子发生效率和附睾精子数量的改变,并用RT—PCR法检测睾丸内精子顶体蛋白酶的表达变化和用酶组织化学方法检测输精管内精子顶体蛋白酶的活性改变。结果:炔雌醇处理的10μg组和25ng组的部分大鼠出现隐睾、睾丸积液和附睾肿物;虽然各处理组大鼠成年后的体重与对照组的相比没有差异,但处理组的双侧睾丸的绝对重量、相对重量和双侧附睾的绝对重量显著下降(P0.01)。第Ⅶ、Ⅷ期生精小管管径大小、小管上皮高度和管腔直径大小除了25ng组与对照组没有明显差异外,其余两组与对照组和25ng组相比,均有明显降低(P0.05)。各处理组大鼠的每日精子产生量和附睾精子总数与对照组相比没有明显差异(P0.05);但处理组的附睾精子数量均比对照组低。给予炔雌醇的剂量越大,顶体蛋白酶呈活性的精子比例就越低,在10μg组、500ng组、25ng组和对照组中该酶呈活性的精子比例分别为54.72±5.87%、64.93±4.06%、73.78±1.87%和81.64±1.55%,各处理组与对照组相比有显著性差异(P0.01)。相对于对照组,10μg组和25ng组大鼠睾丸内顶体蛋白酶的mRNA表达也明显减少(P0.05)。结论:1)新生期雄性大鼠暴露于炔
[Abstract]:Estrogen plays a biological role mainly by binding with estrogen receptor. A large amount of estrogen is produced in the reproductive system of male animals, and the proper concentration of estrogen plays a very important role in the growth and development of male reproduction and the maintenance of normal function. Estrogen deficiency or exposure to high concentrations of estrogen can cause damage to the growth, development and function of reproductive system in male animals, but the mechanism of estrogen in reproductive system is still not well understood. Because of the increasing amount of estrogen and estrogen-active chemicals in the environment, It is urgent to know the action site and mechanism of estrogen on the reproductive system of male animals and whether the environmental estrogens or estrogen-like substances exposed to life of male animals and people will lead to their reproductive structure and positive. Normal functions are adversely affected. The aim of this study was to explore the adverse effects of estrogen exposure on the growth, development and function of the reproductive system in male rats and its mechanism. Methods: from day 2 to day 12, newborn male rats were treated with different doses of 1: 10 渭 g / 2, 500 ng / 2 d and 25ng/2d ethylestradiol (treatment group, control group) with corn oil. After feeding to adulthood, the rats were sacrificed. The morphological structure of testis and epididymis, the spermatogenesis efficiency and the number of epididymal spermatozoa were measured. The expression of acrosin in spermatozoa and the activity of acrosin in spermatozoa were detected by RT-PCR method and enzyme histochemical method respectively. Results: there were cryptorchidism, testicular effusion and epididymal mass in 10 渭 g group treated with ethylestradiol and part of rats in 25ng group, although the weight of adult rats in each treatment group was not different from that of control group, the absolute weight of bilateral testis in treatment group was not different from that in control group.
