细胞因子在雌小鼠内毒素攻击中的变化及其对卵母细胞成熟影响机理的研究

【摘要】:研究背景和研究思路 内毒素(endotoxin)是存在于革兰氏阴性菌外膜中的大分子脂多糖(lipopolysaccharide,LPS)物质,是感染性休克的主要致病因子,而细胞因子如肿瘤坏死因子α(tumor necrosis factor-alpha)、表皮生长因子(epidermal growth factor,EGF)、内皮素-1(endothelin ET-1)、一氧化氮(Nitric oxide,NO)等在介导或保护LPS引起的病理生理学变化中有着非常重要的作用。故而,阻断细胞因子引起的炎症级联反应,,在防治内毒素攻击中具有重要的意义,然而至今尚未寻找到有效的内毒素拮抗剂。 雌性激素是存在于女性体内的正常激素,包括雌激素(以雌二醇—estradiol,E2为代表)和孕激素(以孕酮—progesterone,P为代表)。E_2和P除对生殖系统有影响外,还对机体有重要的保护性作用。人们已经知道,妇女更年期后,易发现骨质疏松和骨折,心血管疾病的发病率也显著升高,即雌性激素对运动系统和心血管系统具有保护作用。现在已经知道,雌性激素对免疫系统、神经系统、消化系统等均发挥调节和保护作用。更重要的是,最近有一系列实验证据证实雌性激素对烧伤、创伤、感染等伤害性刺激造成的免疫系统、神经系统、心血管系统和消化系统等的功能紊乱和器质性损伤具有保护作用。但是,雌性激素的水平随生殖周期的变化而呈现周期性的变化,E_2含量于动情前期逐渐升高,至动情期达高峰,动情间期最低;P含量于动期后期达高峰,动情间期最低。然而,在动情周期的不同阶段行内毒素攻击,对肝、肺等组织造成的病理损伤有无差异尚不清楚,其血浆和卵巢中内源性
[Abstract]:Research background and research ideas Endotoxin (endotoxin) is a macromolecule lipopolysaccharide in the outer membrane of Gram-negative bacteria, which is the main pathogenic factor of septic shock. However, cytokines such as tumor necrosis factor-alpha, epidermal growth factor, endothelin et 1, nitric oxide (no) play an important role in mediating or protecting pathophysiological changes induced by LPS. Therefore, blocking the inflammatory cascade induced by cytokines plays an important role in the prevention and treatment of endotoxin attack. However, no effective endotoxin antagonist has been found so far. Estrogen is a normal hormone in women, including estrogen (estradiolol E2) and progesterone (progesterone P as the representative). Besides affecting the reproductive system, estradiol E 2 and P also play an important protective role in the body. It has been known that after menopause, osteoporosis and fracture are easy to be found, and the incidence of cardiovascular disease is also significantly increased, that is, estrogen has protective effect on the sports and cardiovascular system. It is now known that estrogen regulates and protects the immune system, nervous system, digestive system, etc. More importantly, there has been a recent series of experimental evidence to prove the effects of estrogen on the immune system, nervous system, and other noxious stimuli such as burns, injuries, infections, etc. The dysfunction of cardiovascular system and digestive system and organic damage have protective effect. However, the level of estradiol showed a periodic change with the change of reproductive cycle. The content of E _ 2 increased gradually in the early estrus and reached its peak in estrus, the lowest content of P in estrus reached the peak in the latter stage of estrus and the lowest in estrus. However, it is not clear whether endotoxin attack at different stages of estrous cycle can cause pathological damage to liver, lung and other tissues, and endogenous in plasma and ovary.
