特殊生境中产细菌素乳杆菌的分离、鉴定及益生特性分析

【摘要】:本论文目的在于从健康女性阴道中分离并鉴定产生细菌素、产酸能力强的优势乳杆菌,并对其益生特性作初步研究。分离得到得19 株乳杆菌通过菌落、菌株形态观察以及多项生理生化等常规鉴定,并且通过16S rRNA 基因序列测序鉴定,确定这19 株乳杆菌分属于乳杆菌属(Lactobacillus)内专性同型发酵类型中三个不同的种。其中1-1、1-2、1-3、1-7、51-1、51-4、52-1-1、52-1-2、52-3 共9 株为格氏乳杆菌( Lactobacillus gasseri ), 23-2-1 为唾液乳杆菌  (Lactobacillus.salivarius),23-2-2、23-4、23-5、23-8、32-3、46-1、46-2、46-3、50-3 共9 株为卷曲乳杆菌(Lactobacillus.crispatus)。  这19 株乳杆菌发酵过程中都产生细菌素,除了32-3,51-1,51-4 这3 株菌外,都产生过氧化氢。其36h 发酵液的pH 值都在4 左右。对革兰氏阳性指示菌有强烈的抑制作用,对革兰氏阴性菌以及白色念珠菌均有一定的抑制作用。在杆菌与乳酸球菌以及杆菌之间的拮抗作用实验中,杆菌对同一生境下的乳酸球菌几乎没有抑制作用,杆菌之间,除菌株46-2 对其他的杆菌有抑制作用外,其他的杆菌之间没有抑制作用。  因此这些从人体中分离出来的乳杆菌优势菌群,具有多种维持特殊生境健康的因素,这些作用协同放大,在微环境中会具有强烈的抑制病原菌的功能。本论文所研究的乳杆菌均来源于人体,如应用于人体作为微生态制剂则会具有强大的优势,符合当前女性特殊生境保健的“人性化”潮流。 
[Abstract]:The purpose of this paper is to isolate and identify the dominant Lactobacillus that produce bacteriocin and produce acid from healthy female vagina, and to study its probiotic characteristics. 19 strains of Lactobacillus were identified by colony, morphological observation, physiological and biochemical analysis, and 16s rRNA gene sequencing. The 19 strains of Lactobacillus belong to three different species of homozygous fermentations of Lactobacillus. Among them, 9 strains were Lactobacillus gravis (Lactobacillus gasseri, 23-2-1 was Lactobacillus saliva Lactobacillus, 23-2-2 was Lactobacillus saliva 23-2-2, 23-523-523-532-346-36-46-46-46-3Con 50-3 was Lactobacillus crispatus.Then 9 strains were Lactobacillus crispatusus. All the 19 Lactobacillus strains produced bacteriocin and hydrogen peroxide, except for the three strains of 32-3, 51-1 and 51-4, all of which produced hydrogen peroxide. The pH value of the fermentation broth for 36 h was about 4. It had strong inhibitory effect on Gram-positive indicator bacteria, Gram-negative bacteria and Candida albicans. In the antagonistic experiment between Bacillus lactis and Lactococcus and Bacillus, Bacillus had little inhibitory effect on Lactococcus in the same habitat, but between strains 46-2 had inhibitory effect on other bacilli. There was no inhibition between other bacteria. Therefore, these dominant strains of Lactobacillus isolated from the human body have a variety of factors to maintain the health of special habitats. These effects are co-amplified and have a strong function of inhibiting pathogenic bacteria in microenvironment. The Lactobacillus studied in this paper all come from the human body. If it is applied to the human body as a microecological agent, it will have a strong advantage, which is in line with the current trend of "humanization" in the health care of the female special habitat.
