【摘要】:目的 建立人非小细胞肺癌实验性裸鼠肺部的肿瘤细胞休眠模型,,并验证nm23-H1基因是否具有促进肿瘤细胞休眠的作用。为深入研究肿瘤休眠的机制及治疗提供研究模型。 方法 L9981-nm23-EGFP是nm23-H1基因以真核载体pEGFP质粒转染L9981所构建的人非小细胞肺癌细胞株。L9981-EGFP是转染真核空载体pEGFP质粒的不含nm23-H1基因的人高转移性非小细胞肺癌细胞株。我们以L9981-nm23-EGFP肺癌细胞作为实验组,空载高转移性肺癌细胞株L9981-EGFP作为对照组,分别将两组肿瘤细胞通过尾静脉接种裸鼠,形成实验性非小细胞肺癌的肺部转移模型。8周后断颈法处死全部裸鼠,用免疫组织化学染色(SP)法检测2组裸鼠肺部肺癌细胞的Ki-67增殖指数,TUNEL法检测癌细胞凋亡指数,SP法检测肿瘤组织VEGF的表达,并用流式细胞术检测了裸鼠肺组织的调亡指数和G0/G1期细胞占细胞总数的百分比。 结果 L9981-nm23-EGFP组免疫组化增殖指数、凋亡指数和VEGF的表达均低于高转移性L9981-EGFP组。流式细胞术检测裸鼠肺组织的凋亡指数亦支持免疫组化的结果。实验组G0/G1期细胞占细胞总数的比率
[Abstract]:Objective to establish a tumor cell dormancy model of human non-small cell lung cancer (NSCLC) in nude mice and to verify whether nm23-H1 gene can promote tumor cell dormancy. To provide a model for the further study of the mechanism and treatment of tumor dormancy. Methods L9981-nm23-EGFP is a human non-small cell lung cancer cell line. L9981-EGFP constructed by transfection of nm23-H1 gene with eukaryotic vector pEGFP plasmid. L9981-EGFP is a highly metastatic human non-small cell lung cancer cell line without nm23-H1 gene transfected into eukaryotic vector pEGFP plasmid. L9981-nm23-EGFP lung cancer cells were used as experimental group and non-loaded high metastatic lung cancer cell line L9981-EGFP as control group. Two groups of tumor cells were inoculated into nude mice via tail vein respectively. The lung metastasis model of experimental non-small cell lung cancer (NSCLC) was established. After 8 weeks, all nude mice were killed by cervical amputation. The Ki-67 proliferation index (Ki-67) of lung cancer cells in two groups of nude mice was detected by immunohistochemical staining and the expression of VEGF was detected by Tunel method. The apoptotic index of lung tissue and the percentage of cells in G0/G1 phase in the lung tissue of nude mice were measured by flow cytometry. Results the immunohistochemical proliferative index, apoptosis index and VEGF expression in L9981-nm23-EGFP group were lower than those in high metastatic L9981-EGFP group. The apoptotic index of lung tissue in nude mice by flow cytometry also supported the immunohistochemical results. Ratio of G0/G1 phase cells to total cells in experimental group
