三重脑震荡鼠脑神经元和5羟色胺能神经变化研究

【摘要】:目的:建立三重脑震荡(multidle cerebral concussion,MCC)大鼠模型,观察MCC大鼠大脑皮层、背侧海马、齿状回和脑干网状结构神经元的组织学变化及5羟色胺能神经元、神经纤维变化的规律,探讨与MCC大鼠认知行为障碍相关脑区的神经元和5羟色胺能神经变化,,以了解三重脑震荡鼠认知行为障碍的神经生物学基础。方法:应用金属单摆打击装置复制大鼠MCC模型48只,大鼠随机分为1天、2天、4天、8天、16天和24天6个三重脑震荡组(n=8)。正常对照组(n=8)。用甲苯紫尼氏染色显示神经元,并用光镜对大鼠上述脑区的固缩变性神经元进行计数;用TPH、5—HT抗体免疫组织化学的方法检测大鼠脑干中缝5羟色胺能神经元递质(5—HT)及5—HT合成过程中的限速酶——色氨酸羟化酶(TPH)的表达;用5—HT抗体免疫组化的方法检测大脑皮质、中隔区、伏隔核内侧、丘脑、杏仁体及海马5羟色胺神经纤维的变化。结果:(1)MCC后脑神经元组织化学研究结果:与对照组相比,MCC后1天组大脑前额叶皮质、顶叶皮质和梨状皮质固缩变性神经元丌始增加,2天组达高峰(P<0.05),4天、8天组有所减少,16天组又有所回升,24天组基本恢复正常。颞叶皮质固缩变性神经元在伤后2天达到高峰,4天有所减弱,8天又有回升并接近高峰值,24天组基本恢复正常。海马CA1~CA4和上、下齿状回固缩变性神经元也在伤后1天开始增多,海马CA1~CA4及上齿状回变性神经元数2天达顶峰,下齿状回变性神经元数16天达高峰,至24天与对照组比较无显著性差异。在脑干网状结构脑桥核平面,除16天、24天外,其他各组与对照组比较均有差异(P<0.05)。病变以伤后2天、4天、8天最重(P<0.01),16天有所减轻,24天组基本恢复正常。橄榄核平面病变以伤后2天、4天、8天最重(P<0.01),16天、24天有所减轻,但均未恢复到正常水平。(2)MCC后脑干5—羟色胺能神经元免疫组化研究结果:TPH免疫反应阳性表达在中缝背核、中央上核以伤后1天丌始升高,2天达到高峰,并与对照组相比有显著性差异(p<0.05),24天组基本恢复:与对照组相比,中缝大核以1天组阳性反应最强(p<0.01),中缝苍白核以4天组
[Abstract]:Objective: to establish a rat model of multidle cerebral concussionMCCs and to observe the histological changes of neurons in the cortex, dorsal hippocampus, dentate gyrus and brainstem reticular structure, and the changes of serotonin neurons and nerve fibers in the cerebral cortex, dorsal hippocampus, dentate gyrus and brainstem reticular structure of MCC rats. To investigate the changes of neurons and serotonergic nerves in the brain area associated with cognitive behavioral disorders in MCC rats in order to understand the neurobiological basis of cognitive behavioral disorders in rats with triple concussion. Methods: 48 rat MCC models were established with a metal pendulum device. The rats were randomly divided into two groups: 1 day, 2 days, 4 days, 8 days, 16 days, and 24 days, 6 triple concussion groups, the rats were randomly divided into two groups: 1 day, 2 days, 4 days, 8 days, 16 days, and 24 days, 6 triple cerebral concussion groups. Normal control group (n = 8). The neurons were displayed by toluene violet staining and the pyknotic denatured neurons were counted by light microscope. The expression of serotonin neurotransmitter (5-HT) and the rate-limiting enzyme tryptophan hydroxylase (TPH) during the synthesis of 5-HT in rat brain stem were detected by immunohistochemical method. The changes of serotonin nerve fibers in cerebral cortex, septum, medial nucleus accumbens, thalamus, amygdala and hippocampus were detected by 5-HT antibody immunohistochemical method. Results the results of histochemical study on the posterior cerebral neurons of MCC were as follows: compared with the control group, the prefrontal cortex was found in the prefrontal cortex 1 day after MCC compared with the control group. The pyknotic degeneration neurons of parietal cortex and piriform cortex began to increase at the peak of 2 days group (P < 0.05). The pyknotic denaturation neurons of temporal lobe cortex reached the peak on the 2nd day after injury and decreased on the 4th day, and then recovered on the 8th day and then returned to normal on the 24th day after the injury. The number of hippocampal CA1~CA4 and upper, hypodentate gyrus pyknotic denaturation neurons began to increase 1 day after injury, the number of hippocampal CA1~CA4 and upper dentate degeneration neurons reached its peak on 2 days, and the number of inferior dentate retrograde neurons reached its peak on 16 days. There was no significant difference between 24 days and the control group. The level of pontine nucleus in brain stem reticular structure was significantly higher than that in control group (P < 0.05) except for 16 days and 24 days. The pathological changes were alleviated on the 2nd day, 4th day and 8th day after injury (P < 0.01) or 16 days after injury. In the group of 24 days after injury, the patients returned to normal basically. The most serious lesions in olivary nucleus were observed on the 2nd day, 4th day and 8th day after injury, P < 0.01 and 16 days and 24 days after injury, but they did not return to normal level. The results of immunohistochemical study of serotonin neurons in brain stem showed that the positive expression of TPH immunoreactive reaction in dorsal raphe nucleus was found in the dorsal raphe nucleus. The central superior nucleus increased to the peak at the beginning of 1 day after injury, and there was significant difference between the control group and the control group on the 24th day. Compared with the control group, the nucleus raphe Magnus showed the strongest positive reaction on the 1st day group (p < 0.01), and the pallor nucleus raphe pallidus group on the 4th day group.
