雌二醇和孕酮对小鼠免疫器官淋巴细胞的调节

【摘要】:目的 观察苯甲酸雌二醇、己酸羟孕酮以及二者联用替代疗法对卵巢切除小鼠免疫器官内淋巴细胞数量、形态的变化,以及外周血白细胞、淋巴细胞数量变化,从而探讨苯甲酸雌二醇、己酸孕酮对机体免疫系统的影响。 方法 采用健康雌性昆明小鼠,35天时随机选取48只去势后分成四组,分别注射生理剂量的苯甲酸雌二醇(OVX+EB组)、己酸羟孕酮(OVX+HPC组)或苯甲酸雌二醇、己酸羟孕酮联用(OVX+EB.HPC组)以及麻油(麻油对照组,OVX)。另设一组为保留卵巢空白对照组(INT)。药物注射从小鼠40天开始,每5天注射药物一次,注射30次,共150天。光镜下观察免疫器官内T、B淋巴细胞居住区范围大小的变化;应用MIAS—2000系统分别测定胸腺、脾、淋巴结T、B淋巴细胞居住区细胞密度;电镜下观察各组淋巴结内T、B细胞超微结构的改变。油镜下进行外周血白细胞、淋巴细胞计数。 结果 五组之间进行比较,雌、孕激素对胸腺皮质细胞数量均无影响(P0.05)。OVX+EB和OVX+EB.HPC组与INT组比较均可降低胸腺髓质细胞密度,但二者之间差异在统计学上无意义。OVX+HPC组髓质成熟T细胞密度最高。OVX组髓质细胞密度高于INT。光镜下各组胸腺皮质细胞区域可见OVX+EB和OVX+EB.HPC皮质区域扩大,其余三组不能分辨有何区别。光镜下观察髓质可见,与INT组相比,OVX+EB组髓质区域明显缩小,细胞稀疏,胸腺脂肪化程度最高;OVX+HPC组髓质区域扩大,细胞密集;OVX+EB.HPC组髓质区域大小较INT组无明显改变:OVX组髓质区域明显扩大,脂肪化程度最低。 比较各组脾白髓中央动脉周围淋巴鞘内的T细胞密度和脾小体内B细胞密度出现以下结果:OVX+EB组与INT组相比T细胞密度无差异。OVX+EB.HPC
[Abstract]:Objective to observe the changes of lymphocyte number and morphology in the immune organs of ovariectomized mice by estradiol benzoate, hydroxyprogesterone caproate and their combination therapy, as well as the changes of the number of white blood cells and lymphocytes in peripheral blood of ovariectomized mice. The effects of estradiol benzoate and progesterone hexanoate on immune system were studied. Methods A total of 48 healthy female Kunming mice were randomly divided into four groups after castration for 35 days. Each group was given physiological dose of estradiol benzoate (OVX EB group, OVX HPC group) or estradiol benzoate (Estradiol benzoate). Hydroxyprogesterone caproate combined with OVX EB.HPC group and sesame oil (control group). Another group was used as control group. Drug injection began in mice for 40 days and was injected once every 5 days, 30 times for 150 days. The cell density of thymus, spleen and lymph node were measured by MIAS-2000 system in the living area of T _ (B) B lymphocytes in the immune organs under the light microscope, and the density of the cells in the living areas of the thymus, spleen and lymph nodes were measured by MIAS-2000 system. The ultrastructural changes of TnB cells in lymph nodes were observed under electron microscope. The peripheral blood leukocytes and lymphocytes were counted under oil microscope. Results there was no effect of estrogen and progesterone on the number of thymic cortical cells in the five groups. The density of thymic medulla cells in OVX EB.HPC group and INT group was significantly lower than that in INT group. But the difference between the two groups was statistically insignificant. The density of medullary mature T cells in OVX HPC group was the highest. The medullary cell density in OVX group was higher than that in INT group. Under the light microscope, the OVX EB and OVX EB.HPC cortical regions were enlarged in the thymic cortex of each group, but the other three groups could not distinguish the difference. Compared with the INT group, the medullary area in the OVX EB group was obviously reduced, the cells were sparse, the thymic fat degree was the highest and the medulla area was enlarged in the OVX HPC group. Compared with INT group, the medullary area of OVX EB.HPC group was significantly larger than that of INT group, and the fatty degree was the lowest in OVX group. The density of T cells in peri-lymphatic sheath of central splenic pulposus and B cell density in splenic small body were compared. The results showed that there was no difference in T cell density between the two groups. OVX EB.HPC was not different between the EB group and the INT group.
