女性生殖道新抗菌肽的纯化及鉴定

【摘要】:[背景] 抗菌肽是天然免疫的重要介质,由机体自身基因编码产生,具有抗细菌、抗真菌、抗病毒等生物学活性。女性生殖系统也是天然免疫的重要场所,迄今在女性生殖道中已鉴定出有多种抗菌肽或抗菌蛋白分子存在,例如防御素、溶菌酶、乳铁蛋白、白细胞蛋白酶抑制剂等。是否还存在新的抗菌蛋白或肽分子,目前仍然受到学者们的关注。现今已鉴定出的女性生殖道抗菌肽或抗菌蛋白来源于上皮细胞和/或白细胞,这些天然抗菌分子是否有新的细胞来源,也是令人感兴趣的问题。因此本研究对人子宫内膜粘液和子宫颈粘液的酸溶性提取物进行分离纯化,力图鉴定出新的抗菌肽分子,并对所鉴定出的抗菌肽分子的细胞来源、组织学分布也进行了分析。此外,抗菌肽分子具有固有或诱导表达的特点,因此我们对新鉴定出的分子在临床炎症和非炎症状态下的表达情况进行了分析,以期进一步了解女性生殖道的天然免疫机制,并为其将来用于临床女性生殖道感染防治打下基础。 [方法与结果] 对收集的子宫内膜粘液和子宫颈粘液酸溶性提取物分别进行制备性酸性尿素聚丙烯酰胺凝胶电泳(AU-PAGE),并用电泳琼
[Abstract]:Background: antimicrobial peptides are important mediators of innate immunity. They are encoded by organism's own genes and have biological activities such as antibacterial, antifungal, antiviral and so on. The female reproductive system is also an important place for innate immunity. Up to now, a variety of antimicrobial peptides or antimicrobial proteins have been identified in the female genital tract, such as defensins, lysozyme, lactoferrin, leukocyte protease inhibitors and so on. Whether there are new antimicrobial proteins or peptide molecules is still concerned by scholars. It is also interesting whether these natural antimicrobial molecules are derived from epithelial cells and / or white blood cells. In this study, acid soluble extracts from human endometrial mucus and cervical mucus were isolated and purified in order to identify new antimicrobial peptides, and the cellular origin and histological distribution of identified antimicrobial peptides were also analyzed. In addition, antimicrobial peptide molecules have inherent or inducible expression characteristics, so we have analyzed the expression of newly identified molecules in clinical inflammatory and non-inflammatory states in order to further understand the innate immune mechanism of female genital tract. And lay a foundation for the prevention and treatment of female genital tract infection in the future. [methods and results] Acid urea polyacrylamide gel electrophoresis (AU-PAGEG) was used to prepare acid urea polyacrylamide gel electrophoresis (AU-PAGEN) from the collected endometrial mucus and cervix myxic acid soluble extracts.
