【摘要】:PTEN基因(phosphatase and tension homology deletion on chromosome 10, PTEN)定位在10q23,编码一种具有磷酸酶活性的蛋白,此蛋白与酪氨酸磷酸酶与细胞骨架蛋白tensin和auxilin高度同源。PTEN是一种抑癌基因,PTEN体细胞突变可出现在许多恶性肿瘤的早期和晚期,如前列腺癌、脑胶质瘤、肝癌、肾癌、子宫内膜癌的发生、进展。除了抑癌作用外,PTEN还参与细胞的凋亡、增殖、迁徙和PI3K—AKT信号通路。但近来有研究表明,PTEN与细胞的体积大小有密切关系,尤其是神经元和心肌细胞肥大。文献报道,PTEN的过表达和失活同样都会影响果蝇的眼睛和翅膀体积,敲除PTEN的小鼠出现大头畸形,而在人类大头畸形的患者中检测到PTEN的缺失。影响细胞体积大小的关键因素是细胞内DNA含量;其次,是细胞所处的外界环境;另外一个影响因素是温度。虽然人们一直对细胞体积大小很感兴趣,但其内在的基因调控系统至今仍未阐明。 在人体的正常组织和肿瘤组织中都有许多不同类型的大细胞,如人骨巨细胞瘤中(giant cell tumor of bone,GCT)的破骨样多核巨细胞、卵巢中的
[Abstract]:The PTEN gene phosphatase and tension homology deletion on chromosome 10 (PTEN), located at 10q23, encodes a protein with phosphatase activity. This protein is highly homologous to tyrosine phosphatase and cytoskeletal proteins tensin and auxilin. PTEN somatic mutations may occur in the early and late stages of many malignant tumors, such as prostate cancer, glioma, liver cancer, renal cell carcinoma. The occurrence and progression of endometrial carcinoma. PTEN is involved in cell apoptosis, proliferation, migration and PI3K-AKT signaling. However, recent studies have shown that PTEN is closely related to the size of the cells, especially the hypertrophy of neurons and cardiomyocytes. It has been reported that both overexpression and inactivation of PTEN can affect the size of the eyes and wings of Drosophila melanogaster, and the large head malformation is found in the mice knocking out PTEN, while the deletion of PTEN is detected in the patients with human macrocephaly. The key factor affecting the cell volume is the content of DNA in the cell; secondly, the external environment of the cell; and the other factor is the temperature. Although people have always been interested in the size of the cell, its inherent gene regulation system has not been clarified. There are many different types of large cells in normal and tumor tissues of the human body, such as osteoclast-like multinucleated giant cells in giant cell tumor of human bone, osteoclastic multinucleated giant cells in ovary.
