旋毛形线虫(云南株)感染性及感染机理形态学研究

【摘要】:【目的】 研究旋毛形线虫(云南株)各发育阶段的感染性;探讨雄虫对雌虫生殖能力的影响;通过光镜和电镜对各感染阶段进行观察,从形态学角度阐述其感染机理。 【方法】 138只雄性昆明小鼠,随机分为5组(A、B、C、D_(1-7)、E_(1-6)),A、B、C组以成熟囊包幼虫感染,A组于感染后不同时间剖检,获得不同日龄的成虫和幼虫:B组于感染后5d剖检,确定雌雄性别比例;C组用于计算成熟囊包幼虫的RCI值。取不同日龄的幼虫感D_(1-7);取不同日龄的成虫感染E_(1-5);取单性的5日龄雌虫感染E_6;C、D、E、组均于感染后30天剖检,计算RCI值。取不同感染阶段的幼虫和成虫固定,用光镜和电镜观察其体表、体壁、杆状体、食道、生殖器官。 【结果】 旋毛形线虫(云南株)12、13、14、19日龄幼虫和成熟囊包幼虫的RCI值分别为:49.64±4.02、93.61±4.06、124.34±4.72、153.19±6.20和157.77±3.50;5、10和20日龄成虫的RCI值分别为:154.90±2.62、20.34±4.53和1.18±0.21;5日龄单性雌虫的RCI值为13.77±0.68;11日龄及其以前的幼虫和25日龄及其以后的成虫都不具有感染性。5日成虫的雌雄比例为2.27:1。光镜结果显示:5日龄雌虫子宫内虫卵和幼虫密集,,受精囊和卵巢充盈;15日龄雌虫子宫内虫卵和幼虫稀少,受精囊萎缩;25日龄成虫子宫内未见虫卵和幼虫,受精囊和卵巢结构不清。电镜观察:幼虫体表光滑,头部结构简单;不同感染性的成虫和幼虫体壁结构差异较大,杆细胞中分泌颗粒形态各异,14日龄幼虫杆细胞中可见一种新的梭状的分泌颗粒;各日龄幼虫食道中可见少量非定形物质,而成虫食道
[Abstract]:[objective] to study the infection of nematode spiralis (Yunnan strain) at different developmental stages, to explore the effect of male worms on female reproductive ability, to observe the infection stages by light microscope and electron microscope, and to elucidate the mechanism of infection from the point of view of morphology. [methods] 138 male Kunming mice were randomly divided into 5 groups. The adult and larval larvae of group B were dissected at different times after infection. The adult and larvae of group B were dissected at 5 days after infection. The ratio of female to male sex in group C was used to calculate the RCI value of mature cyst larvae. The larvae of different days were infected with D _ (1-7); the adults of different days were infected with Etix 1-5; the unisexual five-day-old females were infected with E6C ~ (2 +). The RCI values were calculated 30 days after infection. The larvae and adults of different infection stages were fixed, and the surface, wall, rodlike body, esophagus and reproductive organs were observed by light and electron microscopy. [results] the RCI values of nematode nematodes (121213A1419-day-old larvae and mature cysts larvae) were: 49.64 卤4.02293.61 卤4.06124.34 卤4.72153.19 卤6.20 and 157.77 卤3.50510 卤6.20 respectively. The results of light microscope showed that the eggs and larvae were dense in the womb of female worms of 5 days old, the eggs and larvae were rare in the womb of 15 days old female with filling with seminal vesicle and ovary, and there were no eggs and larvae in the womb of 25 days old adult worms with seminal vesicle atrophy. The structure of seminal vesicle and ovary is unclear. Electron microscope observation showed that the surface of larva was smooth, the structure of head was simple, the structure of body wall of adult and larva with different infection was different, and a new kind of secretory granules could be found in bar cells of 14-day-old larva. A small amount of amorphous substances were found in the esophagus of each day old larva, while the adult oesophagus
