【摘要】:碱性成纤维细胞生长因子和转化生长因子β1对体外培养幼兔关节软骨细胞增殖的影响 目的 碱性成纤维细胞生长因子和转化生长因子β1与软骨细胞的生长、分化十分密切,因此在软骨组织工程研究中受到了普遍的关注。本文研究碱性成纤维细胞生长因子(bFGF)和转化生长因子β1(TGF-β1)在单一和联合应用时,对体外培养幼兔关节软骨细胞增殖的影响,探讨软骨组织工程中能够有效促进软骨细胞增殖的两种生长因子的浓度组合方式。 方法 体外多代培养的幼兔关节软骨细胞(第2、4、6、8代)分别以bFGF(1,5,10ng/ml)、TGF-β1(0.1,0.5,1ng/ml)及其不同浓度间两两组合的联合刺激组为实验组,以常规培养组为对照组,通过CellTiter96(?)AQueous单溶液细胞增殖分析法测定各代细胞在生长因子(及其联合)作用下的增殖效应。 结果 单独应用时,1ng/ml的TGF-β1为促增殖的最佳效应浓度;bFGF在第2、4、6代细胞中的促增殖最佳浓度为10ng/ml,在第8代细胞中的最佳浓度为5ng/ml;联合应用时,当TGF-β1为低浓度(0.1ng/ml)或中浓度(0.5ng/ml)联合bFGF时,高浓度bFGF(10ng/ml)与之组合促增殖效果最佳;当高浓度TGF-β1(1ng/ml)与bFGF联合时,对早期(第2、4代)细胞,TGF-β1(1ng/ml)+bFGF(10ng/ml)为最佳的促增殖浓度组合,而在后期(第6、8代),,TGF-β1(1ng/ml)
[Abstract]:Effects of basic fibroblast growth factor and transforming growth factor beta 1 on the proliferation of rabbit articular chondrocytes in vitro
Objective basic fibroblast growth factor (GGF) and transforming growth factor beta 1 (TGF - beta 1) are closely related to the growth and differentiation of chondrocytes. Therefore, it is widely concerned in the study of cartilage tissue engineering. In this paper, basic fibroblast growth factor (bFGF) and transforming growth factor beta 1 (TGF- beta 1) were studied in a single and combined application to the culture in vitro. The effects of two kinds of growth factors on chondrocyte proliferation in cartilage tissue engineering were discussed.
Methods the chondrocytes of young rabbit joint in vitro (2,4,6,8) were treated with bFGF (1,5,10ng / ml), TGF- beta 1 (0.1,0.5,1ng / ml) and the combination of 22 combinations of different concentrations as the experimental group. The normal culture group was used as the control group. The cells of each generation were determined by the CellTiter96 (?) AQueous single solution cell proliferation analysis. Proliferation effect under the action of long factor (and its combined).
Results the optimal concentration of 1ng / ml TGF- beta 1 was the best effect concentration for promoting proliferation, and the best concentration of bFGF in 2,4,6 cells was 10NG / ml, and the best concentration in the eighth generation cells was 5ng / ml. When the TGF- beta 1 was low concentration (0.1ng / ml) or medium concentration L) is the best combination to promote proliferation. When the high concentration of TGF- beta 1 (1ng / ml) is combined with bFGF, the optimal proliferation concentration combination for the early (2,4) cells, TGF- beta 1 (1ng / ml) +bFGF (10NG / ml), and in the later period (1).
