甲型肝炎病毒BL-08株的研究

【摘要】: 本论文在成功地进行了毒株的分离,适应性培养以及鉴定之后,从传代的细胞中提取出病毒BL-08的基因组RNA,再采用逆转录-聚合酶链式反应(RT-PCR)对甲肝病毒BL-08进行扩增、克隆、测序得基因组序列nt25-7476。再将HAV BL-08株的基因序列与其它株型进行比较,得到了以下的结果:全长核苷酸序列同源性与MBB株最高,为98.8%;与L-A-1株次之为98.0%,与HM175野毒株的同源性为95.0%。而在氨基酸同源性方面却显示出相反的结果,MBB株和HM175株与BL-08序列的氨基酸同源性分别为98.4%和98.5%。 另外,本论文还着重对甲肝病毒BL-08株的5’NTR区进行研究。pHAV/7质粒是由PH.D.Suzanne Emerson提供的,以pEGM载体为骨架,带有HAV HM175株全基因组序列的质粒,全长10358bp。本实验在原有的pHAV/7的基础上,将有内部核糖体结合位点(IRES)的5’NTR区域与BL-08株做相应位置的置换,然后将其和pHAV/7进行比较,从而分析单独的5’NTR区对于HAV病毒生长周期的影响。
[Abstract]:After successful isolation, adaptive culture and identification of the virus strain, the genomic RNAs of virus BL-08 were extracted from the passage cells and then amplified and cloned by reverse transcription-polymerase chain reaction (RT-PCR). The genomic sequence nt25-7476 was sequenced. By comparing the gene sequence of HAV BL-08 strain with other strains, the following results were obtained: the homology of full-length nucleotide sequence with MBB strain was 98.8, that with L-A-1 strain was 98.0 and that with HM175 field strain was 95.0. The results of amino acid homology showed that the amino acid homology between MBB strain and HM175 strain was 98.4% and 98.5% with BL-08 sequence, respectively. In addition, the 5'NTR region of hepatitis A virus (BL-08) strain was studied in this paper. The plasmid pHAV / 7 was provided by PH.D.Suzanne Emerson. The plasmid containing the whole genome sequence of HAV HM175 strain was pEGM vector and the whole length of plasmid was 10358bp. On the basis of the original pHAV/7, the 5'NTR region with internal ribosomal binding site (IRES) was replaced with the BL-08 strain, and then compared with pHAV/7. The effect of single 5'NTR region on the growth cycle of HAV virus was analyzed.
