【摘要】:钩体为广泛流行的人兽共患病——钩体病的病原体。目前,对钩体的致病及免疫分子机理的认识不甚清楚。赖型钩体56601株及哥本哈根株基因组测序表明,约60%的钩体预测基因的功能未知。因此,钩体的重要致病及免疫相关基因功能急需加强研究。 invA基因存在于致病的赖型钩体56601株及哥本哈根株中,信息分析表明,该基因与巴尔通体内的与编码侵袭能力相关蛋白IalA的基因等具有很高的相似性;以PCR扩增试验也证实,invA基因存在于强毒赖型钩体017株中,但不存在于无毒钩体Patoc Ⅰ株中。提示invA基因很可能为致病相关基因,同时,也很可能是一个免疫侯选基因及用于钩体诊断的目的基因。 本研究选择赖型钩体017株中invA基因为目标,PCR扩增出含完整读码框的invA基因,与原核表达载体pET32a(+)连接构建重组表达载体pETINVA,在大肠杆菌BL21(DE3)中表达出分子量约为35kDa的InvA融
[Abstract]:Leptospira is the pathogen of zoonosis-leptospirosis. At present, the pathogenesis of leptospirosis and the mechanism of immune molecules are not well understood. Genomic sequencing of 56601 and Copenhagen Leptospira strains showed that about 60% of Leptospira predictive genes were unknown. Therefore, the important pathogenicity and immune-related gene function of Leptospira urgently need to be studied. InvA gene exists in 56601 strains of Leptospira and Copenhagen strain. There was a high similarity between the gene and the gene encoding invasion ability related protein IalA in Balton, and the PCR amplification test also confirmed that the inva gene existed in the highly virulent Leptospira Leptospira strain 017, but not in the non-toxic Leptospira Patoc 鈪,
