【摘要】: 目的:寻找一种简单、可靠的方法来建立并评估豚鼠膀胱逼尿肌不稳定(detrusor instability,DI)模型。观察与Cajal间质细胞(interstitial cells of Cajal, ICCs)相似的一种细胞在DI膀胱逼尿肌中的形态、分布变化,探讨其与DI发病机理的相关性。 方法:成年雌性豚鼠经会阴部不完全尿道结扎,6周后行膀胱穿刺充盈性膀胱测压,观察膀胱压力、容量变化及逼尿肌不稳定的发生情况。根据检测结果,将建立的实验豚鼠模型分为DI组和逼尿肌稳定(detrusor stabilization,DS)组。另设正常对照组,采用组织冰冻切片、酪氨酸蛋白激酶受体(receptor tyrosine kinase,KIT)单克隆抗体间接免疫荧光检测技术观察逼尿肌中ICCs样细胞的形态、分布情况。 结果:①实验组逼尿肌不稳定发生率35.9%,对照组无发生,其中DI组膀胱最大压力和最大容量较对照组显著增加(p0.01)。②在成年豚鼠膀胱的逼尿肌中存在有ICCs样细胞,该细胞走向与逼尿肌肌束平行,主要分布于逼尿肌肌束边缘和逼尿肌肌束中,呈梭形双极细胞。与对照组比较,DI组ICCs样细胞数量明显增多。 结论:①经会阴部尿道结扎、膀胱穿刺充盈性膀胱测压是建立及评估豚鼠膀胱出口梗阻引起逼尿肌不稳定模型的一种简单、可靠的方法。②KIT单克隆抗体间接免疫荧光检测技术是有效观察膀胱ICCs样细胞的免疫组织化学实验方法。③DI膀胱中ICCs样细胞增多提示其可能导致逼尿肌自发收缩活动增加,可能就是理论上存在于膀胱的起搏细胞,可能是DI发生的重要因素之一。
[Abstract]:Objective: to find a simple and reliable method to establish and evaluate detrusor instabilities DI model in guinea pigs. The morphology and distribution of a cell similar to Cajal interstitial cell (interstitial cells of Cajal in DI bladder detrusor muscle were observed, and the relationship between it and the pathogenesis of DI was discussed. Methods: six weeks after incomplete urethral ligation of perineum, the bladder pressure, volume and detrusor instability of adult female guinea pigs were measured by bladder puncture and filling bladder manometry. According to the test results, the guinea pig model was divided into DI group and detrusor stabilization DS group. The morphology and distribution of ICCS-like cells in detrusor muscle were observed by indirect immunofluorescence assay with monoclonal antibody against tyrosine protein kinase receptor (receptor tyrosine kinaseKit) in normal control group. Results the incidence of detrusor instability was 35.9in the experimental group, but not in the control group. The maximal pressure and volume of bladder in DI group were significantly higher than those in the control group (p0.01) .2 ICCs like cells were found in the detrusor muscle of adult guinea pig bladder. The direction of the cell was parallel to the detrusor bundle, mainly distributed in the edge of the detrusor fascicle and in the detrusor bundle, with fusiform bipolar cells. Compared with the control group, the number of ICCs like cells in DI group was significantly increased. Conclusion it is a simple method to establish and evaluate the model of detrusor instability caused by bladder outlet obstruction in guinea pigs. The reliable method. 2KIT monoclonal antibody indirect immunofluorescence assay is an effective method for observing ICCs like cells in bladder. The increase of ICCs like cells in bladder suggests that the increase of ICCs like cells in bladder may lead to the increase of detrusor spontaneous contraction activity. May be theoretically present in the bladder pacemaker cells, may be one of the important factors of DI.
