基因芯片技术在成年男性精子基因表达谱研究中的应用

【摘要】:不孕不育是全世界关注的课题。目前每100对已婚夫妇中,就有10对夫妇存在不孕不育问题。其中男性因素占到了50%。也就是说,男性精子质量不佳,已影响到人类的繁衍生息。 引起精子质量不佳的原因很多,长期以来人们进行了多方面的研究,但病因和发病机制都不是十分明确,临床治疗效果也不尽如人意。男性精子质量不佳,从而引发的男性不育,成为困扰临床的难题之一。从分子水平探明影响精子质量的因素,以便采取有针对性的诊断、治疗措施,提高精子质量,提高人类生育力,具有重要的理论和现实意义。 遗传学的发展提示我们“人类的疾病或多或少与基因有关”,男性精子质量不佳也应是如此。因此,我们试图从基因的水平来解释男性精子质量不佳的原因,以期能够对临床采取有针对性的、更有效的诊断、治疗措施,提供理论上的支持和指导。从基因的水平阐述疾病的致病机制,可以有两个途径。一是从基因组DNA的完整性、正确性入手,研究精子基因组DNA是否存在DNA碱基序列的大片段缺失,或出现DNA碱基的某些位点处的突变、缺失等。另一方面则是着眼于基因的表达,因为人类有3×10~(10)个碱基对,但仅仅表达3万个左右的蛋
[Abstract]:Infertility is a worldwide concern. Currently, for every 100 married couples, 10 couples have infertility problems. Male factors account for 50% of the total. In other words, male sperm quality is poor, has affected human reproduction. There are many reasons for the poor quality of sperm. For a long time, people have carried out many studies, but the etiology and pathogenesis are not very clear, and the effect of clinical treatment is not satisfactory. The poor quality of male sperm, which leads to male infertility, has become one of the clinical problems. It is of great theoretical and practical significance to identify the factors affecting sperm quality at molecular level in order to take targeted diagnosis and treatment measures to improve sperm quality and improve human fertility. The development of genetics suggests that "human diseases are more or less genetically related", as should the poor quality of male sperm. Therefore, we try to explain the reasons for the poor quality of male spermatozoa from the gene level, in order to provide theoretical support and guidance for clinical diagnosis and treatment. There are two ways to explain the pathogenesis of disease at the gene level. The first is to study whether there are large deletion of DNA base sequence, mutation and deletion at some sites of DNA base in sperm genomic DNA from the integrity and correctness of genomic DNA. On the other hand, it's about gene expression, because humans have 3 脳 10 ~ (10) base pairs, but only about 30,000 eggs.
