骨形态发生蛋白和转化生长因子β诱导成骨的协同作用

【摘要】:本实验应用不同浓度的盐酸胍和尿素反复溶解沉淀蛋白质的方法从牛骨中分离制备骨形态发生蛋白(BMP),以及应用酸醇法从猪血小板中提取制备转化生长因子β(TGF-β),并将制备的样品植入小鼠股部肌肉内进行诱导成骨实验,旨在探索BMP诱导成骨的最佳方式。实验结果如下: (1) 相同条件下,使用载体比无载体具有更强的诱导能力,合适载体的使用会使诱导实验取得更好的效果; (2) BMP与TGF-β联合使用比单独使用BMP具有更好的诱导成骨效果,表明BMP的诱导只是启动过程,TGF-β可促进诱导物进一步分化增殖; (3) 从载体组的诱导使用可以看出,新骨形成后,其碱性磷酸酶的活性降低,表明此时新生细胞表达减弱,合成有机质降低;而从钙的变化可看出,此时更多的是无机质的形成,即骨中钙盐的沉积; (4) 通过细胞生物学实验同样证明BMP与TGF-β联合使用比单独使用BMP具有更好的诱导成骨效果,而且可以验证骨的修复再生来源于间充质干细胞。 综上所述,BMP诱导成骨的最佳条件是,使用合适的材料作为载体,同时与其他生长因子联合应用。
[Abstract]:In this experiment, bone morphogenetic protein (BMP) was isolated from bovine bone with different concentrations of guanidine hydrochloride and urea, and transforming growth factor 尾 (TGF- 尾) was extracted from porcine platelets by acid alcohol method. The samples were implanted into the femoral muscles of mice for osteoinduction. The aim of this study was to explore the best method of osteogenesis induced by BMP. The experimental results are as follows: (1) under the same conditions, the use of the carrier has a stronger induction ability than the non-carrier. (2) the combination of BMP and TGF- 尾 has better osteogenic effect than that of BMP alone. The results indicated that BMP induction was only a priming process, and TGF- 尾 could promote the further differentiation and proliferation of the inducer. (3) from the induction of carrier group, we can see that after the formation of new bone, The activity of alkaline phosphatase was decreased, which indicated that the expression of new cells decreased and the synthesis of organic matter decreased, but the change of calcium showed that the formation of inorganic substance was more likely at this time. (4) the combined use of BMP and TGF- 尾 also proved that BMP and TGF- 尾 could induce osteogenesis better than BMP alone. And it can be proved that bone repair and regeneration come from mesenchymal stem cells. To sum up, the best condition of BMP osteoinduction is to use suitable materials as carrier and combined with other growth factors.
