兔中性粒细胞防御素对A549细胞IL-8基因表达的影响

【摘要】:【背景】脊椎动物防御素(defensins)是小分子的阳离子抗菌多肽,它们广泛分布于上皮细胞和吞噬细胞中。人和兔中性粒细胞的总蛋白中5%以上为防御素,它们除了具有直接的抗菌功能外还能作为免疫活性分子作用于上皮细胞和免疫细胞,参与天然免疫和获得性免疫的调节。另外它们还可作为信号分子调节细胞信号转导过程,,促进细胞的增殖、分化等。探索防御素的非抗菌功能是目前防御素研究的重点方向之一,本研究从mRNA和蛋白水平初步探讨了兔中性粒细胞防御素(rabbits neutrophils antimicrobial peptides,NPs)对人Ⅱ型肺泡上皮细胞A549 IL-8表达的影响及其作用机制。 【方法与结果】向家兔腹腔注射1%酪蛋白350ml,构建兔腹腔无菌性炎症动物模型。收集腹腔炎性渗出液,离心后弃上清,用双蒸水破红细胞,离心后超声破碎白细胞,再次离心收集上清。上清经Bio-Gel P-10低压色谱凝胶层析柱分离纯化后,用RP-HPLC法经C18柱进一步纯化,
[Abstract]:Background: vertebrate defensin (defensins) is a cationic antibacterial polypeptide of small molecules, which is widely distributed in epithelial cells and phagocytes. More than 5% of the total proteins in human and rabbit neutrophils are defensins. In addition to their direct antimicrobial function, they can act as immune active molecules in epithelial cells and immune cells, and participate in the regulation of innate and acquired immunity. In addition, they can also act as signal molecules to regulate cell signal transduction and promote cell proliferation and differentiation.
