一种新型CpG ODN对乙肝疫苗增效作用的研究

【摘要】:重组乙型肝炎表面抗原(HBsAg)疫苗是在世界范围内广泛应用的预防乙型肝炎病毒的有效疫苗,但在我国约10%的人群对其反应低下或无反应,因此研制一种高效的乙型肝炎疫苗已成为当务之急。 CpG ODN 是人工合成的含有CpG 的单链寡聚脱氧核苷酸,具有强效免疫增强作用。CpG ODN 可分为A、B、C 三类,其中B 类CpG ODN(CpG-B)能有效刺激B 细胞介导的体液免疫应答,可望成为增强HBsAg 免疫效果的理想分子佐剂。 为了寻找能够有效增强HBsAg 免疫原性的CpG ODN,我们进行了大规模的新型CpG ODN 筛选工作,发现一种新型的CpG ODN(CpG-684)能够刺激淋巴细胞增殖、活化、刺激IFN-γ的产生,且能明显提高HBsAg刺激小鼠产生抗体的能力。说明这种新型CpG ODN 有可能成为乙肝疫苗的高效佐剂。
[Abstract]:Recombinant hepatitis B surface antigen (HBsAg) vaccine is an effective vaccine for the prevention of hepatitis B virus in the world, but about 10% of the people in our country have low response or no response to it. Therefore, it has become an urgent task to develop a highly effective hepatitis B vaccine.
CpG ODN is a synthetic single strand oligo deoxynucleotides containing CpG, which can be divided into three classes of A, B, C, which can be divided into three classes of A, B, and C. The B class CpG ODN (CpG-B) can effectively stimulate the humoral immune response mediated by the cells. It is expected to be a molecular adjuvant to enhance the immune effect.
In order to find CpG ODN which can effectively enhance the immunogenicity of HBsAg, we carried out a large-scale new CpG ODN screening work. It was found that a new type of CpG ODN (CpG-684) can stimulate lymphocyte proliferation, activation, stimulate the production of IFN- gamma, and can obviously improve the ability of HBsAg stimulates mice to produce antibodies. This new CpG ODN is available. It can be an efficient adjuvant for hepatitis B vaccine.
