B淋巴细胞刺激因子治疗性疫苗的研究

【摘要】:B淋巴细胞刺激因子(B Lymphocyte Stimulator,BLyS)是1999年由瑞士和美国科学家发现的肿瘤坏死因子(Tumor Necrosis Factor,TNF)家族的一个新成员。该分子含有285个氨基酸,属于Ⅱ型跨膜蛋白,编码区位于染色体13q32-34。BLyS主要由T细胞和树突状细胞等髓系来源的细胞分泌。BLyS以膜结合以及可溶性两种方式存在于机体中,两者均具有生物学活性。 迄今为止,已经发现了三种受体可以与BLyS结合,分别为BCMA、TACI和BAFF-R,由于缺少信号转导序列,它们被划归为Ⅲ型膜蛋白。其中前两者也可以与其他的分子结合,而BAFF-R则为BLyS特异性的受体,这些受体仅位于B细胞膜表面,使得BLyS的生物学活性——刺激B淋巴细胞的成熟以及增殖,有了高度的选择性。 研究表明,BLyS在B细胞的存活和功能调节方面发挥着重要作用,它可以延长B细胞存活时间、促进其增殖,从而导致B细胞增生和自身免疫狼疮样变化的出现。BLyS的过度表达是发生自身免疫性疾病,尤其是系统性红斑狼疮的重要因素之一。但是,对于BLyS的作用机制还不明了。
[Abstract]:B lymphocyte stimulating factor (B Lymphocyte timulatorus (BLyS) is a new member of the tumor necrosis factor (Tumor Necrosis) family discovered by scientists in Switzerland and the United States in 1999. The molecule contains 285 amino acids and belongs to type II transmembrane protein. The coding region is located in the chromosome 13q32-34.BLyS secreted mainly by myeloid cells such as T cells and dendritic cells. BLyS exists in the organism in two ways: membrane binding and soluble. Both have biological activity.
