健康人肠道大肠埃希菌耐药性及其耐药基因播散机制的研究

【摘要】:目的 测定北京地区健康人肠道大肠埃希菌耐药水平和特征;研究整合子和耐药基因盒在健康人群肠道大肠埃希菌中携带情况及分布特点,探讨耐药基因盒与细菌耐药表型的关系;了解整合子在不同基因型菌株中的分布,以及整合子在耐药性水平传递中的作用和分子机制。 方法 利用全自动细菌鉴定/药敏分析仪,对从健康人肠道分离的150株大肠埃希菌进行鉴定,14种抗生素敏感性测定和产ESBLs判读,另外7种抗生素用纸片扩散法进行敏感性测定;对两种药物敏感性方法进行平行实验比较结果符合率;采用PCR方法检测细菌携带Ⅰ、Ⅱ类整合子的情况,,通过限制性内切酶分析,纯化测序,将不同大小Ⅰ类整合子的DNA序列在GeneBank中搜索,确定其可变区基因盒的种类;利用脉冲场凝胶电泳(PFGE)对大肠埃希菌进行分子分型;通过质粒接合试验研究Ⅰ类整合子
[Abstract]:Objective to determine the level and characteristics of drug resistance of Escherichia coli in healthy people in Beijing area. To study the distribution of integron and drug resistance gene box in Escherichia coli in healthy population, to explore the relationship between drug resistance gene box and bacterial phenotype, and to understand the distribution of integron in different genotypes of Escherichia coli. And the role and molecular mechanism of integron in drug resistance level transmission. Methods automatic bacterial identification / drug sensitivity analyzer was used. Fifteen strains of Escherichia coli isolated from healthy human intestines were tested for their susceptibility to 14 antibiotics and ESBLs interpretation, and 7 antibiotics were detected by disk diffusion method. The coincidence rate of the two drug sensitivity methods was compared by parallel experiments. PCR method was used to detect bacteria carrying class 鈪
