外周血单个核细胞在不同条件下HSP70形成的研究

【摘要】:目的:热休克蛋白(hot shock protein ,HSP)又称作分子伴侣,是所有细胞能在应激情况下所产生的一组蛋白质。根据分子量的大小分为 HSP20、HSP70、HSP90 和大分子 HSP 家族。HSP70 是 HSP 家族中最重要的一种。HSP70 被认为同时与新生的多肽链结合部分,HSP70 作为防止蛋白质的聚集及错误折叠的媒介。HSP70 与机体的生理作用密切相关,参与细胞内的许多功能,如蛋白质折叠、运转、成熟及降解。目前对 HSP 的研究成为热点,但 HSP 的确切作用还仍未了解,特别是介质对热休克蛋白形成的影响研究未见报道。本试验的目的是通过人外周血单个核细胞不同时间内在不同 pH 和不同介质中 HSP70 形成的研究,探讨影响热休克蛋白形成的因素。 方法:抽取无发烧、无炎症的健康志愿者外周血,应用密度梯度法分离单个核细胞,分别在不同 pH 的 AB 型血清和不同介质(1640 培养液、AB 型人血清、PBS 缓冲液)中 42℃水浴中热休克 0.5、1 小时,用免疫组化法测定热休克蛋白 70 的形成。 结果:外周血单个核细胞在不同pH的AB型血清中42℃热休克0.5、1 小时,热休克蛋白 70 的表达与未作热休克处理的空白比较有明显差异,P0.01。外周血单个核细胞在 AB 血清中(pH7.35)42℃热休克0.5、1 小时热休克蛋白 70 的形成有明显差异,P0.01。热休克 0.5小时的表达明显高于 1 小时;酸性(pH6.95)AB 血清中 42℃热休克 0.5、1 小时热休克蛋白 70 的形成无明显差异,P0.05;碱性(pH7.65)AB血清中 42℃热休克 0.5、1 小时热休克蛋白 70 的形成无明显差异,P 0.05。 外周血单个核细胞在不同介质中 42℃热休克 0.5、1 小时,热休克蛋白 70 的表达与空白比较有显著性差异,P0.01。在磷酸盐缓冲液中42℃热休克0.5、1小时,热休克蛋白70的形成有明显差异,P0.01,0.5 小时的表达高于 1 小时;1640 培养液中 42℃热休克 0.5、1 小时,热休克蛋白 70 的表达差异不明显,P0.05。 结论:(1)外周血单个核细胞热休克蛋白 70 形成的量与应激源数量密切相关,多种应激源刺激热休克蛋白质 70 形成的量比单一应激源刺激形成的快。(2)碱性条件下外周血单个核细胞也可产生热休克蛋白。(3)外周血单个核细胞热休克蛋白的形成与细胞所处的介质密切相关。
