维甲酸和刺猬因子促进骨髓基质细胞向运动神经元发育的实验研究

【摘要】:本实验共分为两部分研究维甲酸(RA)和刺猬因子(Shh)单独和联合诱导对促进骨髓基质细胞(BMSCs)向运动神经元(MN)发育的作用。第一部分使用本实验室制神经干细胞培养基(专利号:ZL02134314.4)培养骨髓基质细胞(BMSCs),掌握BMSCs的原代培养和纯化方法,观察细胞生长、发育和分化情况,并通过检测神经干细胞标记Nestin和MN发育过程重要标记蛋白Hb9,研究了BMSCs的增殖、分化生物学特点。第二部分研究依诱导剂不同分四组(空白组、RA组、Shh组和RA+Shh组)诱导BMSCs,,观察骨髓源性神经干细胞(BMSCs-D-NSCs)的生长增殖及分化情况,检测Nestin和Hb9标记表达,并结合发育神经生物学的知识探讨BMSCs-D-NSCs进一步分化为运动神经元的潜力。 第一章 小鼠骨髓基质细胞的培养及生物学特性观察 目的 研究BMSCs取材、原代培养方法,观察细胞在特制培养基的生长、增殖和形态变化状况,掌握BMSCs-D-NSCs体外培养要点和生长生物学特性,获得稳定纯化的BMSCs-D-NSCs,为后续诱导分化实验奠定基础。
[Abstract]:This experiment was divided into two parts to study the effects of (RA) and (Shh) on the development of (BMSCs) motoneuron (MN) in bone marrow stromal cells. The first part was to use the culture medium of neural stem cells (patent no.: ZL02134314.4) to culture bone marrow stromal cells (BMSCs),) to master the primary culture and purification methods of BMSCs, and to observe the growth, development and differentiation of the cells. The biological characteristics of proliferation and differentiation of BMSCs were studied by detecting neural stem cell labeled Nestin and Hb9, an important marker in the development of MN. The second part was to observe the growth, proliferation and differentiation of bone marrow derived neural stem cells (BMSCs-D-NSCs) by BMSCs, induced by BMSCs, and to detect the expression of Nestin and Hb9 markers in four groups (control group, RA group, Shh group and RA Shh group). Combined with the knowledge of developmental neurobiology, the potential of BMSCs-D-NSCs to differentiate into motor neurons was discussed. The first chapter: culture and biological characteristics of mouse bone marrow stromal cells The study of BMSCs material, The primary culture method was used to observe the growth, proliferation and morphological changes of the cells in the special medium, to master the main points of BMSCs-D-NSCs culture in vitro and to obtain the stable and purified BMSCs-D-NSCs, which laid the foundation for the subsequent induction and differentiation experiment.
