狂犬病毒脂质体疫苗的研究

【摘要】:目前广泛应用的狂犬病疫苗为VERO细胞纯化灭活疫苗,以氢氧化铝做为佐剂,需免疫五次,且保护效力较低。本文制备了狂犬病毒脂质体疫苗,以期减少免疫次数,并且提高疫苗的保护效力。 我们以氢化大豆磷脂、胆固醇、十八胺为原料制备空白脂质体,以海藻糖为冻干保护剂。采用冻干重建法(REV-DRV)制备狂犬病毒脂质体疫苗。采用冻干重建法制备的狂犬病毒脂质体疫苗形态成圆形或椭圆形,粒径分布均匀,平均粒径在12.25μm左右,包封率在65%以上。本品无刺激性,安全试验、过敏试验均合格。以脂质体为载体的狂犬疫苗同铝佐剂疫苗相比,可提高效力3倍,中和抗体脂质体疫苗组明显高于对照铝佐剂组,两组之间相比有显著性差异。采用冻干重建法制备的狂犬病毒脂质体疫苗可同时增强细胞免疫和体液免疫,提高免疫效力,有望减少免疫针次。
[Abstract]:The widely used rabies vaccine is purified inactivated VERO cell vaccine. Aluminum hydroxide as adjuvant needs to be immunized five times and the protective efficacy is low. The rabies virus liposome vaccine was prepared in order to reduce the immunization times and improve the protective effect of the vaccine. Blank liposomes were prepared from hydrogenated soybean phospholipid, cholesterol and octadecylamine, and trehalose was used as lyophilization protectant. Liposome vaccine against rabies virus was prepared by freeze-dried reconstruction (REV-DRV). The rabies virus liposome vaccine prepared by freeze-drying reconstruction method is round or elliptical in shape. The particle size distribution is uniform, the average particle size is about 12.25 渭 m, and the encapsulation efficiency is over 65%. This product has no irritation, safety test and allergy test. Compared with aluminum adjuvant vaccine, rabies vaccine with liposome as carrier could increase the potency by three times. The neutralizing antibody liposome vaccine group was significantly higher than that of control aluminum adjuvant group, and there was significant difference between the two groups. The liposome vaccine of rabies virus prepared by freeze-dried reconstruction can enhance both cellular and humoral immunity and improve the immune efficacy.
