冈比亚按蚊触角气味结合蛋白基因AgOBP48表达载体的构建及在大肠杆菌中的表达

【摘要】:疟疾是严重危害人体健康的疾病之一,引起人类疟疾的所有疟原虫都是按蚊属昆虫传播的,,非洲冈比亚按蚊高度嗜吸人血,是恶性疟原虫极其高效的传播媒介,所以危害尤为严重,昆虫都是依靠嗅觉发现寄主的,嗅觉在按蚊的搜索寄主行为中起关键作用,昆虫能够感受空气中的挥发性气味分子,并依此作为寻偶、觅食和寻找产卵场所的信息。而昆虫触角感受器中的气味结合蛋白被认为是运输气味分子的受体,是昆虫专一性识别外界气味物质的第一步生化反应,对于昆虫与外界进行信息交流具有重要意义,本论文开展了对冈比亚按蚊的气味结合蛋白AgOBP48基因的研究。 利用英国洛桑研究所惠赠的含有AgOBP48基因的PET-20b(+),用NdeⅠ/EcoRⅠ双酶切取得目的基因片段,将目的片断和原核表达载体pET-28a(+)经NdeⅠ/EcoRⅠ双酶酶切重组到表达型质粒中并转化入原核细胞中。表达序列测定结果表明:冈比亚按蚊气味结基因AgOBP48的成熟蛋白阅读框全长536bp,编码172个氨基酸残基,预测分子量和等电点分别为19kDa和5.00。 本研究将冈比亚按蚊AgOBP48基因整合入原核表达载体中表达,得到的重组蛋白在其N-端融合有His·Tag纯化标记,为下一步蛋白的纯化,功能研究奠定基础。
