STAg联合IFN-γ和蜂胶鼻内免疫小鼠诱导的抗弓形虫感染保护性免疫应答

【摘要】:目的本研究首先探讨STAg 分别联合不同剂量IFN-γ、蜂胶鼻内免疫对小鼠的保护作用及IFN-γ、蜂胶作为佐剂鼻内免疫抗弓形虫感染的最佳剂量。再比较IFN-γ、蜂胶两种佐剂最佳剂量以及两种佐剂混合鼻内免疫辅助STAg增强机体抗弓形虫攻击的能力,并探索两种佐剂联合应用的免疫效果。 方法确定IFN-γ、蜂胶最佳剂量的实验方法:分别将70 只、60 只5~6 周龄BALB/c小鼠随机分为5个组,用20μg STAg分别联合不同剂量IFN-(γ250U,500U,1000U,2000U)、蜂胶(5μg,10μg,20μg, 40μg)鼻内免疫小鼠2 次,间隔14 天,末次免疫后第10 天,用RH 株弓形虫速殖子4×104 个/只灌胃攻击,逐日观察小鼠健康存活情况。攻击后第29 天处死全部小鼠,分离计数脑、肝组织速殖子;计数肠系膜淋巴结(MLN)和脾T 淋巴细胞;ELISA 法检测小鼠粪便、肺冲洗液弓形虫特异性IgA 和血清IgG 抗体水平。 比较IFN-γ、蜂胶佐剂作用的实验方法:将5~6 周龄雌性BALB/c 小鼠60 只随机分为4 个组,每组15 只,分别用20μg STAg、20μg STAg+40μg蜂胶、20μg STAg+1000U IFN-γ以及20μg STAg+40μg 蜂胶+1000U IFN-γ鼻内免疫小鼠2 次,间隔14 天,末次免疫后第10 天,用RH 株弓形虫速殖子4×104 个/只灌胃攻击,逐日记录小鼠体重,观察小鼠存活情况。攻击后第43 天处死全部小鼠,分离计数脑、肝组织速殖子;计数IEL、PP 结和脾T 淋巴细胞,免疫细胞化学(ICC)法检测其T 细胞亚群水平;ELISA 法检测小鼠粪便、鼻咽冲洗液、肺冲洗液、阴道冲洗液和血清中弓形虫特异性IgA 和血清IgG 抗体水平。 结果随佐剂IFN-γ剂量的增加,小鼠存活率有升高趋势,1000U IFN-γ剂量组小鼠存活率最高达93%;脑、肝速殖子数呈下降的趋势, 其中STAg
[Abstract]:Objective to investigate the protective effect of STAg combined with different doses of IFN- 纬, propolis nasal immunization on mice and the optimal dose of IFN- 纬 and propolis as adjuvant in nasal immunity against Toxoplasma gondii infection. The best dosage of IFN- 纬, two adjuvants of propolis and two adjuvants combined with STAg were compared to enhance the ability of body to resist Toxoplasma gondii attack, and to explore the immune effect of the two adjuvants combined with the two adjuvants.
