小鼠lrp的克隆表达及功能的初步研究

【摘要】:脂多糖(LPS)是革兰氏阴性细菌外膜的主要组成部分。它能通过对抗抗菌剂来保护细菌的细胞壁。脂多糖是包含疏水脂质和亲水多糖两部分的异聚物。在溶菌后,胞壁的LPS被释放入血液循环系统,通过刺激单核细胞、巨噬细胞、中性粒细胞、血小板及内皮细胞而激活免疫系统。脂多糖还能绑定到血浆白蛋白形成复合物,增加LPS的毒性以及LPS对膜受体的亲和力。LPS刺激免疫细胞后导致炎症因子的释放,如:细胞活素类、化学增活素、酶类、花生四烯酸类物质、粘着剂以及自由基,这些物质对炎症反应过程起着重要的作用,而且能够诱导包括脓毒性休克在内的病理生理学过程。因此,研究脂多糖应答分子对解决LPS引起的包括脓毒性休克在内的一系列病理生理学过程具有重要的意义。 为此,,课题组致力于探讨LPS刺激后应答基因的功能,尤其是一些新发现基因的功能。lrp是一种LPS应答基因。系本课题组柴玉波博士采用基于PCR的改良消减杂交技术,构建LPS刺激后人牙髓细胞差异表达基因文库,从中筛选出的新基因,GenBank登录号为AF143740。后来在GenBank又有更为完整的
[Abstract]:Lipopolysaccharide (LPS) is a major component of the outer membrane of Gram-negative bacteria. It protects the cell walls of bacteria by anti-antimicrobial agents. Lipopolysaccharide is an heteropolymer consisting of hydrophobic lipids and hydrophilic polysaccharides. After bacteriolysis, LPS from the cell wall is released into the circulatory system, activating the immune system by stimulating monocytes, macrophages, neutrophils, platelets and endothelial cells. Lipopolysaccharide also binds to plasma albumin to form complexes that increase the toxicity of LPS and the affinity of LPS to membrane receptors. LPS stimulates the release of inflammatory cytokines such as cytokines, chemoactivins and enzymes. Arachidonic acids adhesives and free radicals play an important role in inflammatory processes and induce pathophysiological processes including septic shock. Therefore, the study of lipopolysaccharide responsive molecules plays an important role in solving a series of pathophysiological processes, including septic shock caused by LPS. Therefore, the research group is devoted to study the function of the response gene after LPS stimulation, especially the function of some newly discovered genes. Lrp is a kind of LPS response gene. By using improved subtractive hybridization based on PCR, Dr Chai Yubo, our research group, constructed the differentially expressed gene library of human dental pulp cells stimulated by LPS.
