重复性经颅磁刺激生物效应及其机制的研究

【摘要】: 目的 重复性经颅磁刺激刺激(repetitive transcranial magnetic stimulation, rTMS)是一种新型非创伤性的神经科学研究工具,并可能成为多种精神疾病的治疗手段。目前对rTMS的抗抑郁(样)作用的性别差异研究较少,而且不同作用方式的rTMS对学习和记忆的影响尚不明确,另外rTMS的作用机制可能与神经可塑性有关。因此,本实验研究重复性经颅磁刺激的生物效应,重点研究rTMS的抗抑郁样作用及其长期rTMS抗抑郁样作用的性别差异;并考察rTMS对学习和记忆的影响,研究短期高频率rTMS对空间学习和记忆过程的影响,长期低频率rTMS对学习和记忆的影响;同时研究长期rTMS对大鼠海马Bcl-2蛋白表达的影响。 方法 Wistar雄、雌性大鼠接受rTMS连续作用10 d(15 Hz,1.0 Tesla,共2000次刺激)。以强迫游泳实验观察抗抑郁作用及其性别差异。Wistar雄性大鼠接受rTMS连续作用5 d,分别在第1 d和第5 d rTMS结束后接受旷场实验。 本研究中,Wistar雄性大鼠接受高频率rTMS(10 Hz,1.0 Tesla,200μs)300次刺激。使用Morris水迷宫实验,考察训练前或后给予rTMS处理对空间参考记忆和工作记忆的影响。
[Abstract]:Objective repetitive transcranial magnetic stimulation (repetitive transcranial magnetic stimulation, rTMS) is a new non-traumatic neuroscience tool and may be used as a therapeutic tool for various mental disorders. At present, there are few studies on the gender difference of antidepressant (like) effects of rTMS, and the effects of rTMS on learning and memory are not clear. In addition, the mechanism of rTMS may be related to neural plasticity. Therefore, we studied the biological effects of repetitive transcranial magnetic stimulation (TMS), focusing on the antidepressant effect of rTMS and the gender difference of long-term antidepressant effect of rTMS. The effects of rTMS on learning and memory, the effects of short-term high frequency rTMS on spatial learning and memory, long-term and low frequency rTMS on learning and memory, and the effects of long-term rTMS on the expression of Bcl-2 protein in hippocampus of rats were studied. Methods Wistar male and female rats were treated with rTMS for 10 days (15 Hz,1.0 Tesla, for 2000 times). The antidepressant effect and its gender difference were observed by forced swimming test. Wistar male rats were treated with rTMS for 5 days, and the open field test was performed on the 1st and 5th day after rTMS. In this study, Wistar male rats were stimulated with high frequency rTMS (10 Hz,1.0 Tesla,200 渭 s) 300 times. Morris water maze experiment was used to investigate the effects of rTMS treatment on spatial reference memory and working memory before or after training.
