自然衰老SD大鼠肾阳虚模型的研究

【摘要】:目的: 自然衰老SD大鼠经常作为肾阳虚证动物模型应用于补肾阳药物的研究开发以及肾阳虚与衰老关系的研究中,但在应用时存在着月龄的差异,所使用的大鼠月龄主要集中在24月龄到30月龄之间,究竟哪个月龄段的大鼠,相对来说更适宜作为肾阳虚证动物模型,文献上没有明确的指出。本课题使用筛选的肾阳虚证动物模型评价标准及温补肾阳的中药对不同月龄自然衰老SD大鼠进行初步的验证和治疗,并根据不同月龄的自然衰老SD大鼠的检测结果以及反证疗效的差异,筛选出较适宜性别和月龄的自然衰老SD大鼠的肾阳虚模型,为中医证候动物模型规范化研究及抗衰老的药物研究提供支持和帮助。 方法:本实验分为三部分进行 实验一:自然衰老SD大鼠肾阳虚的观察 选取雄性的青年大鼠作为对照组,雄性的自然衰老大鼠作为模型组通过观察两组动物的症状、体征以及血清皮质醇(COR)、三碘甲状腺原氨酸(T_3)、甲状腺素(L_4)、睾酮(T)/雌二醇(E_2)、脏器指数、病理改变等指标来评价自然衰老雄性大鼠是否具有肾阳虚证的实质。 实验二:连续月龄的自然衰老SD大鼠肾阳虚损程度的比较 取连续三个月龄的衰老早期的大鼠进行实验,实验结束后取血、取材对比它们之间的血清皮质醇(COR)、三碘甲状腺原氨酸(L_3)、甲状腺素(L_4)、睾酮(T)/雌二醇(E_4)、脏器指数,通过对比研究寻找出差异的最小间隔期为下一次试验提供依据。 实验三:肾阳虚证自然衰老SD大鼠月龄的筛选 按照实验二的结果作为实验周期将实验分成三个不同的月龄段进行,首先取一定数量的24月龄大鼠随机分为两组,一组为空白组,另一组为药物治疗反证组,空白组每天灌胃两毫升无菌水,药物组灌胃两毫升右归丸水煎剂,期间观察两组的症状体征,实验结束后采集血清血浆—60℃冰箱保存,第二次取26月龄的自然衰老SD大鼠、最后取28月龄的自然衰老SD大鼠进行相同过程的实验。 结果: 1.实验一:自然衰老SD雄性大鼠具有肾阳虚的症状体征,检测结果基本符合所筛选肾阳虚的指标。 2.实验二:用所筛选的血清学指标检测连续月龄的自然衰老SD雄性大鼠时,出现差异的时间至少间隔一个月。 3.实验三:26到28月龄段为最佳自然衰老肾阳虚模型。 结论:
[Abstract]:Objective: natural aging SD rats are often used as an animal model of kidney-yang deficiency syndrome in the research and development of kidney-yang drugs and the relationship between kidney-yang deficiency and aging. The age of the rats is mainly between 24 months and 30 months old. The objective of this study was to use the selected animal model of kidney-yang deficiency syndrome and the Chinese medicine to warm up the kidney-yang to verify and treat the natural aging SD rats of different months of age. According to the test results of natural aging SD rats of different ages and the difference of curative effect, the kidney yang deficiency model of natural aging SD rats with suitable gender and month age was selected. To provide support and help for the standardization of animal models of TCM syndromes and the research of anti-aging drugs. Methods: this experiment was divided into three parts. One was the observation of kidney-yang deficiency in natural aging SD rats. Young male rats were selected as the control group. Male natural aging rats were used as model group to observe the symptoms, signs and serum cortisol (COR), triiodothyronine (T _ S _ 3) and thyroxine (L _ 4) in the two groups. Testosterone (T) / estradiol (E _ 2), viscera index and pathological changes were used to evaluate the essence of kidney yang deficiency in natural aging male rats. Experiment 2: comparison of the degree of deficiency of Kidney Yang in naturally aged SD Rats aged 3 months. Blood samples were taken from the rats at the early stage of aging, and the blood samples were taken after the end of the experiment.
