脑源性神经营养因子在体外培养大鼠胚脑皮质神经元低氧适应中的保护作用

【摘要】:第一部分:大鼠胚脑皮质神经元细胞的原代培养、鉴定和缺氧模型的建立 目的:进行大鼠胚脑皮质神经元细胞培养和鉴定,并建立大鼠胚脑皮质神经元细胞培养的缺氧模型。 方法:(1)运用胰蛋白酶和DNase Ⅰ联合酶消化法,选择合适孔径的筛网过滤,,并以L-多聚赖氨酸作为神经元体外培养的生长基质,进行大鼠胚脑皮质神经元的体外培养;(2)运用免疫荧光组织化学方法检测神经元特异的抗TubulinβⅢ单克隆抗体和神经胶质细胞特异的抗GFAP多克隆抗体分别进行神经元细胞和神经胶质细胞的鉴定,检测体外培养大鼠胚脑皮质神经元细胞的纯度;(3)运用国产厌氧培养箱在37℃含85%N_2、5%CO_2、10%H_2混合气体环境下建立大鼠胚脑皮质神经元细胞的缺氧模型。 结果:(1)通过体外培养获得了纯度达95%以上的大鼠胚脑皮质神经元细胞;(2)运用神经元特异的抗TubulinβⅢ单克隆抗体和神
