大鼠下丘脑内NOS与ER双染神经元的表达

【摘要】: 目的:探讨一氧化氮合酶和雌激素受体在下丘脑诸核团的分布及共存,为揭示雌激素与一氧化氮之间内在联系提供形态学证据。方法:对30只SD雌性大鼠下丘脑切片,采用还原型尼克酰胺腺嘌呤二核苷酸脱氢酶(NADPH-d)组织化学方法并结合免疫组织化学技术,观察大鼠下丘脑内NOS阳性神经元、ER阳性神经元以及NOS/ER双染神经元的形态及分布。同时结合图像分析技术,计算下丘脑各核团内NOS阳性神经元、ER阳性神经元以及NOS/ER双染神经元的数目和双染神经元所占的比例。结果:①NOS阳性神经元主要分布在下丘脑室旁核、视上核、下丘脑外侧区和室周核。②ER阳性神经元在下丘脑诸核团的表达不及NOS阳性神经元广泛,主要分布在下丘脑视上核、室旁核和下丘脑外侧区,此外在室周核和大脑皮层也见到少量散在分布的ER阳性神经元。③NOS与ER双染神经元主要分布在下丘脑的室旁核、视上核、下丘脑外侧区及室周核。其它区域可见到散在分布的双染神经元。视上核双染神经元主要位于视上核背内侧和背外侧部。室旁核的双染神经元主要分布在小细胞部的腹内侧区。在下丘脑外侧区,双染神经元分布较广。室周核可见零星分布的双染神经元。结论:1在大鼠下丘脑视上核、室旁核、下丘脑外侧区及室周核均有NOS与ER双染神经元分布。2 NOS与ER双染神经元主要集中分布在视上核的背内侧和背外侧部及室旁核小细胞部腹内侧区,在下丘脑外侧区分布较广但比较分散,室周核呈散在分布。
[Abstract]:Aim: to investigate the distribution and coexistence of nitric oxide synthase (NOS) and estrogen receptor (ER) in hypothalamic nuclei, and to provide morphological evidence for revealing the relationship between estrogen and nitric oxide (no). Methods: the NOS positive neurons in the hypothalamus of 30 SD female rats were observed by using reduced nicotinamide adenine dinucleotide dehydrogenase (NADPH-d) histochemistry and immunohistochemical technique. Morphology and distribution of ER positive neurons and NOS/ER double staining neurons. At the same time, the number and proportion of NOS positive neurons, ER positive neurons and NOS/ER double staining neurons in each nucleus of hypothalamus were calculated with image analysis technique. Results: 1NOS positive neurons were mainly distributed in the paraventricular nucleus of hypothalamus, supraoptic nucleus, lateral hypothalamic area and periventricular nucleus. The expression of 2ER positive neurons in the nuclei of hypothalamus was not as wide as that of NOS positive neurons, but mainly in the supraoptic nucleus of hypothalamus. The paraventricular nucleus and the lateral hypothalamus were also found in the paraventricular nucleus and lateral hypothalamus, and a few scattered ER positive neurons were found in the periventricular nucleus and cerebral cortex. The double staining neurons of 3NOS and ER were mainly distributed in the paraventricular nucleus, supraoptic nucleus, lateral hypothalamic area and periventricular nucleus of the hypothalamus. Scattered double-stained neurons can be seen in other regions. The double staining neurons of supraoptic nucleus were mainly located in the dorsomedial and dorsolateral part of supraoptic nucleus. The double-staining neurons in the paraventricular nucleus were mainly located in the ventromedial region of the small cells. In the lateral hypothalamus, double staining neurons are widely distributed. Scattered double-stained neurons were found in the periventricular nucleus. Conclusion: 1 in the supraoptic nucleus and paraventricular nucleus of the hypothalamus of rats, The distribution of NOS and ER double staining neurons in lateral hypothalamic area and periventricular nucleus. 2 NOS and ER double staining neurons were mainly distributed in dorsomedial and dorsolateral part of supraoptic nucleus and ventromedial area of small cell part of paraventricular nucleus. The lateral hypothalamus was widely distributed, and the periventricular nucleus was scattered.
