A型肉毒毒素对电场刺激及SP引发的胃离体平滑肌收缩作用及机理研究

【摘要】: 目的:观察A型肉毒毒素(botulinum toxin A,BTX-A)对大鼠幽门、胃窦离体平滑肌条自发性收缩和电场刺激(electrical field stimulation,EFS)、P物质(substance P,SP)引发的幽门、胃窦离体平滑肌收缩的影响,并探讨其机制。 方法:选取健康的Sprague-Dawley大鼠,体重200~250g,雌雄不拘。实验前禁食24h,饮水不限。实验时击头部致昏后取幽门、胃窦平滑肌各一条,置于37℃Krebs液的恒温平滑肌槽中,肌槽内持续供给95%O_2和5%CO_2的混合气体,肌条的一端固定在肌槽底部的玻璃弯钩上,另一端固定在张力传感器上,肌条被悬于两个银电极之间,肌条长轴与电极长轴平行,电极与电刺激器相连。肌条在1g的前负荷下温育,随机分为对照组(control,n=12)、电场刺激组(electrical field stimulation,EFS,n=12)、EFS+BTX-A组(n=12)、BTX-A组(n=12)、SP组(n=12)、SP+BTX-A组(n=12)、SP+NK_1受体拮抗剂([d-arg~1,d-phe~5,d-trp~(7.9),leu~(11)]-substance P)组(n=12)。在自发性收缩的条件下,分别加入BTX-A(10 U/ml)、SP(1μM/L)、NK_1受体拮抗剂(1μM/L)及EFS(频率16Hz,电压35V,波宽0.5ms持续时间60-s),Biolap 420E生物机能实验系统记录胃肌条收缩情况。 结果:1.EFS引发幽门平滑肌张力增强(P<0.01)和振幅增强(P<0.05),EFS引发胃窦平滑肌张力及振幅增强(P<0.01);2.BTX-A抑制EFS引发的幽门、胃窦平滑肌张力和振幅(P<0.01);3.BTX-A导致幽门、胃窦平滑肌张力和振幅降低(P<0.01);4.SP引发幽门平滑肌振幅增强(P<0.05),张力无显著性差异(P>0.05),SP引发胃窦平滑肌张力增强(P<0.01)和振幅增强(P<0.01);5.BTX-A抑制SP引发的幽门平滑肌振幅(P<0.01),不抑制SP引发的幽门平滑肌张力(P>0.05),BTX-A抑制SP引发的胃窦平滑肌张力和振幅(P<0.01);6.NK_1受体拮抗剂抑制SP引发的幽门平滑肌张力(P<0.01),不抑制SP引发的幽门平滑肌振幅(P>0.05),,NK_1受体拮抗剂抑制SP引发的胃窦平滑肌张力(P<0.01)及振幅(P<0.01)。 结论:1.EFS增强幽门、胃窦平滑肌自发性收缩;2.BTX-A抑制幽门、胃窦平滑肌自发性收缩;3.BTX-A抑制EFS引发的幽门、胃窦平滑肌收缩;提示BTX-A可能通过抑制内源性递质释放或对直接电场刺激肌肉产生的收缩的抑制,从而抑制胃平滑肌的收缩。4.SP增强幽门、胃窦平滑肌收缩;5.BTX-A抑制SP引发的幽门、胃窦平滑肌收缩;6.NK_1受体拮抗剂抑制SP引发的幽门、胃窦平滑肌收缩,提示SP可能通过和平滑肌上受体结合发挥作用,使肌肉收缩增强,而NK_1受体拮抗剂则可能通过拮抗SP与受体结合使肌肉收缩减弱。BTX-A可能通过抑制突触囊泡与突触前膜融合、胞吐,从而抑制递质释放发挥作用,也可能对SP与突触后膜上相应受体结合具有一定的影响作用,导致对SP引发的幽门、胃窦离体平滑肌收缩的抑制。

Effects of isolated gastric antrum smooth muscle contraction and its mechanism were discussed. Methods: healthy Sprague-Dawley rats were selected, weighing 200 g or 250 g, male and female. Fasting for 24 hours before the experiment, drinking water is unlimited. The pyloric pylorus and antral smooth muscle were taken after the head fainted and placed in the isothermal smooth muscle groove of 37 鈩
