激活素受体相互作用蛋白2a全长基因克隆及其与ActRIP1-3的比较研究

【摘要】:激活素作为TGF-β超家族的细胞增殖、分化因子,其作用的多面性和组织学特异性,与激活素作用的细胞内信号传导差异有关。在激活素信号传导中,激活素受体相互作用蛋白(Activin receptor-interacting protein,ActRIP)是一组新发现的信号传导蛋白。本研究在克隆新的ActRIP家族蛋白——ActRIP2a基因基础上,进一步比较研究ActRIP2a与ActRIP1-3组织分布、细胞内表达格局以及生物学活性的差异。 本研究克隆了新的激活素受体相互作用蛋白2a(ActRIP2a)全编码基因,cDNA全长1008bp,编码153个氨基酸残基,与ActRIP2仅在编码区第15和99位氨基酸不同,ActRIP2a的PDZ功能区与ActRIP1的蛋白结合功能区PDZ5存在33%同源性,与ActRIP3的ActR Ⅱ结合功能区具有96%同源性,但与ActRIP3的C末端无同源性。哺乳细胞双杂交系统证实ActRIP2a可与ActR ⅡA相互作用,免疫沉淀实验进一步证实ActRIP2a可与ActR ⅡA稳定结合,Pull-down实验结果表明ActRIP2a通过其N末端PDZ样功能区与天然ActR ⅡA相互作用,免疫沉淀实验表明ActRIP2a可以形成同源双聚体蛋白,但与ActRIP2不能构成异源双聚体。 Northern杂交显示ActRIP1、2、3 mRNA表达存在组织学分布上的差异,RT-PCR检测结果显示,ActRIP2a mRNA在大脑组织、垂体、睾丸中表达较高,胰腺、卵巢组织中表达较弱,而在其它组织中未见明显表达;ActRIP1 mRNA在脑干、大脑、小脑、垂体、下丘脑、睾丸、肾上腺组织中有表达,其他组织内未见表达;而sActRIP1(short ActRIP1)在心肌及脾脏组织内未见表达,其他组织均有不同程度表达。ActRIP2及ActRIP3则在多种组织内均有表达。免疫组织化学染色结果也显示,ActRIP2在小鼠各组织几乎均有着色;ActRIP1在肺、胰腺及睾丸的曲精细管内几乎没有染色反应,而在其他组织有不同程度着色;小鼠脑
[Abstract]:Activin, as a cell proliferation and differentiation factor of TGF- 尾 superfamily, has many facets and histological specificity, which is related to the difference of intracellular signal transduction induced by activin. Activin receptor interaction protein (Activin receptor-interacting protein,ActRIP) is a newly discovered signal transduction protein in activin signal transduction. On the basis of cloning ActRIP2a gene, a new ActRIP family protein, the differences of tissue distribution, intracellular expression pattern and biological activity between ActRIP2a and ActRIP1-3 were further studied. In this study, a novel activin receptor interaction protein 2a (ActRIP2a) gene was cloned. The total length of cDNA is 1008bpand encodes 153 amino acid residues, which is different from that of ActRIP2 only at the 15th and 99th amino acids in the coding region.
