蛋白酶激活受体在人嗜碱性粒细胞中的表达及对其组胺释放的影响

【摘要】:背景与目的: 蛋白酶激活受体(Protease Activated Receptors,PARs)属于G蛋白偶联受体 家族成员,分布在多种细胞上。目前在人和小鼠共发现4种PARs,分别为PAR-1、 PAR-2、PAR-3和PAR-4。PARs可被多种蛋白酶激活,参与机体多种生理与病 理生理过程。目前,越来越多的研究显示PARs在炎症和变态反应中起着重要的 作用。 嗜碱性粒细胞是一种重要的免疫细胞。细胞表面表达高亲和力IgE受体 (FcεRI),并在相应的过敏原作用下引起受体交联,导致细胞脱颗粒从而释放多种 介质,包括组胺、IL-4、IL-13、白三烯、前列腺素E_2等,通过这些介质直接作 用或者活化其它细胞从而参与炎症和变态反应等过程。 虽然PARs在炎症与变态反应中的作用已成为研究热点,但迄今为止,嗜碱 性粒细胞的PARs相关研究尚未见报道。4种PARs在嗜碱性粒细胞上的表达情 况如何,PARs对嗜碱性粒细胞的激活有无调节作用?以上种种问题尚待解答。 PARs对人嗜碱性粒细胞组胺释放的影响的研究将有助于人们对PARs功能的了 解,并且对进一步阐明嗜碱性粒细胞在变态反应发生中的作用具有重要意义。 材料和方法: 用TRIZOL试剂提取人嗜碱性粒细胞系KU812总RNA,然后进行RT-PCR
[Abstract]:Background & objective: protease-activated receptor (Protease Activated Receptors,PARs) is a member of the G-protein coupled receptor family and is distributed in a variety of cells. At present, four kinds of PARs, PAR-1, PAR-2,PAR-3 and PAR-4.PARs, were found to be activated by a variety of protease in human and mouse, respectively. Participate in a variety of physiological and pathological physiological processes. At present, more and more studies have shown that PARs plays an important role in inflammation and metamorphosis. Basophil is an important immune cell. High affinity IgE receptor (Fc 蔚 RI),) is expressed on the cell surface and is cross-linked with the corresponding allergen, resulting in the degranulation of cells and the release of a variety of mediators, including histamine. IL-4,IL-13, leukotriene, prostaglandin E _ 2 and so on, through these mediums are directly used or activated other cells to participate in inflammation and allergic reaction and other processes. Although the role of PARs in inflammation and hypersensitivity has become a hot topic, so far, PARs related studies on basophil have not been reported. How about the expression of four kinds of PARs on basophil? does PARs regulate the activation of basophil? The above questions remain unanswered. The study of the effect of PARs on the histamine release of human basophilic granulocytes will contribute to the understanding of the function of PARs, and it will be of great significance to further clarify the role of basophil in the pathogenesis of allergic reaction. Materials and methods: total RNA, of human basophilic granulocyte KU812 was extracted with TRIZOL reagent and then RT-PCR was performed.
