两株甲真菌rDNAITS序列分析及系统形态学研究

【摘要】:甲真菌病是由真菌引起的一种慢性、顽固、防碍美容的常见传染性疾病。本文对从甲病患者病甲中分离得到的菌株,进行个体发育学研究,弥补了当前甲真菌研究中对其分类地位确定的不完善之处,并探讨了某些分子生物学技术在甲真菌分类中的应用价值。 1.通过体外模型实验确定了菌株Fh_4和菌株Dh_3是引起甲部感染的病原真菌。 2.通过不同生长阶段菌体的DNA提取结果,丝状真菌DNA的提取应选在其对数生长期为好。 根据系统分类研究中的18S rDNA序列和rDNAITS序列(ITS1、5.8S rDNA、ITS2)与相关属、种的序列构建的系统发育树、序列相似性研究,菌株Fh_4的分类地位应是半知菌亚门(Fungi Imperfecfi)—球壳孢目(Sphaeropsidales)—球壳孢科(Sphaeropsidaceae)—茎点霉属(Phoma)。菌株Dh_3的分类地位应是半知菌亚门(Fungi Imperfecti)—丛梗孢目(Moniliales)—丛梗孢科(Monilialiaceae)—枝顶孢属(Acremonium)。 3.通过对菌株Fh_4和Dh_3进行了较为系统的形态学研究可知:菌株Fh_4的适宜的生长条件是28℃,pH 6.5-7.0;菌落从白色变为红色再变为暗黄色;菌丝无色,具隔膜,分枝状生长;能产生关节菌丝;分生孢子单胞红色,椭圆形;分生孢子器球形,黑褐色,顶端有孔口,孢子成熟后从孔口逸出;没有发现有性阶段。菌株Dh_3适宜的生长条件是:28℃,pH 6.0-7.0;在察氏培养基上菌落淡红色;营养菌丝匍匐生长,分枝、光滑、具隔膜,宽1.5μm,分生孢子梗散生,瓶梗直接从菌丝上长出,直立锥形,多单生,产孢瓶体细长28~38μm,圆柱状,分生孢子单个循序产生,形成链状,孢子椭圆形,表面光滑,4.4~5.2×1.4~2.0μm,未见厚垣孢子;未发现Dh_3产生有性孢子器。 4.通过以上研究及相关资料和检索表可知:菌株Dh_3应是枝顶霉属中的Acremonium egyptiacum菌株Fh_4属于茎点霉属,由于茎点霉属菌种以前都是根据真菌感染的植物而命名,使得属中的同种异名现象很严重,而且到现在未见有关茎点霉属的真菌能引起甲真菌病的报道,建议给菌株Fh_4新的名称。
[Abstract]:Onychomycosis is a chronic, persistent, beauty-impaired, common infectious disease caused by fungi. In this paper, the individual development of strains isolated from patients with nail disease was studied, which made up for the imperfections in the classification status of onychomycetes in the current research. The application value of some molecular biological techniques in the classification of onychomycetes was also discussed. 1. Strain Fh_4 and strain Dh_3 were identified as pathogenic fungi causing infection of part A by model experiment in vitro. 2. The extraction of DNA from filamentous fungi by DNA extraction at different growth stages should be selected in the logarithmic growth period of filamentous fungi. Based on the phylogenetic tree and sequence similarity of 18s rDNA sequence and rDNAITS sequence (ITS1,5.8S rDNA,ITS2) and related genera and species in phylogenetic classification, The taxonomic status of strain Fh_4 should be (Fungi Imperfecfi)-(Sphaeropsidales)-(Sphaeropsidaceae)-(Phoma). The taxonomic status of strain Dh_3 should be (Fungi Imperfecti)-(Moniliales)-(Monilialiaceae)-(Acremonium). 3. The morphology of strain Fh_4 and Dh_3 was studied systematically.
