外周5-羟色胺2A受体在角叉菜胶诱发的痛敏和炎症中的作用

【摘要】:本研究采用行为学和免疫组织化学等生物学技术,,探讨外周5—羟色胺2A(5-HT_(2A)受体)在致炎物质角叉菜胶(carrageenan)诱发的痛觉过敏和炎症中的作用。 大鼠足趾部注射5-HT_(2A)受体拮抗剂酮色林(ketanserin)剂量依赖性地(0.5、3和5μg)抑制了角叉菜胶诱发的痛觉过敏、炎症和脊髓腰4~5节段背角c-fos蛋白的表达。但是,5-HT_(1A)受体拮抗剂WAY-100635却无此抑制作用。不同浓度(0.5%,1%和3%)的酮色林局部贴剂剂量依赖性地抑制了由高岭土和角叉菜胶诱发的关节炎性疼痛和炎症,而对非炎症侧没有任何影响,表明这种作用是通过外周而不是中枢神经系统。 本研究表明外周5—羟色胺参与一般慢性和关节炎中疼痛和炎症的形成,而且这种作用是通过外周5—羟色胺2A受体介导的。
[Abstract]:The purpose of this study was to investigate the role of peripheral 5-hydroxytryptamine 2A (5-HT _ (2A) receptor) in hyperalgesia and inflammation induced by carrageenan (carrageenan) by means of behavioral and immunohistochemical techniques. A dose-dependent injection of 5-HT2A receptor antagonist ketelin (ketanserin) (0.5, 3 and 5 渭 g) inhibited carrageenan-induced hyperalgesia, inflammation and the expression of c-fos protein in the dorsal horn of the lumbar 4 and 5 segments of the spinal cord. However, 5-HT _ (1A) receptor antagonist WAY-100635 had no inhibitory effect. Different concentrations (0.5%, 1% and 3%) of ketoselin local patches dose-dependently inhibited inflammatory pain and inflammation in the joints induced by kaolin and carrageenan, but had no effect on the non-inflammatory side. This suggests that this effect is through the peripheral rather than central nervous system. This study suggests that peripheral serotonin is involved in the formation of pain and inflammation in general chronic and arthritis, and this effect is mediated by peripheral serotonin 2A receptor.
