广西马尔尼菲青霉菌微卫星多态性

【摘要】: 目的研究广西马尔尼菲青霉菌(PM)的基因多态性,比较PM竹鼠分离株和临床分离株的基因多态性差异。 方法采用氯化苄提取法提取基因组DNA。用微卫星标记多态性分析法对40株竹鼠分离PM和31株临床分离PM菌株进行基因多态性分析。 结果①40只广西野生银星竹鼠自然携带PM阳性率100%。②微卫星标记多态性分析从71株PM中检测到53个微卫星类型,并具有较高区分度(D=0.990)。微卫星Ⅰ,Ⅱ,Ⅲ分别检测到21,10,和9个等位基因。③广西银星竹鼠体内寄生PM各地区间的遗传相似系数为0.1217~0.4464之间。④广西竹鼠PM与广西马尔尼菲青霉病患者(PSM)分离PM遗传相似系数为0.8689.⑤广西野生银星竹鼠PM与广西PSM患者分离PM各地区间的遗传相似系数为0.0604~0.8418之间。 结论广西马尔尼菲青霉菌存在的基因多态性;广西银星竹鼠自然携带之PM存在地域上的基因多态性;引起人类PSM的病原菌可能与广西野生竹鼠自然携带PM有密切的相关性。

Methods Genomic DNA. was extracted by benzyl chloride extraction method. The genetic polymorphisms of 40 strains of PM isolated from bamboo mice and 31 strains of clinical isolates of PM were analyzed by microsatellite marker polymorphism analysis.
