抗多种致病菌的特异性卵黄免疫球蛋白的制备研究

【摘要】:目的:探讨一种新的获得IgY多克隆抗体的方法和效果。方法:用三种常见的细菌病原体金黄色葡萄球菌(SA)、大肠埃希氏菌(EC)、铜绿假单胞菌(PA)制备复合抗原,并经过灭活后免疫来亨蛋鸡产生IgY多克隆抗体。从免疫后的蛋中提取出IgY多克隆抗体,ELISA法测IgY抗体特异性及浓度滴度。SA、EC、PA三种标准菌株体外抑制实验检测IgY抗体抑菌效果。结果:用SA、EC、PA复合抗原成功地免疫来亨蛋鸡产生IgY多克隆抗体,ELISA法检测抗体浓度滴度为100,000;IgY抗体明显抑制SA、EC、PA三种标准菌株的生长。结论:SA、EC、PA三种细菌复合抗原可以刺激来亨蛋鸡产生高浓度滴度抗体IgY,该抗体能明显抑制SA、EC、PA三种标准菌株的生长。
[Abstract]:Objective: to explore a new method for obtaining polyclonal antibody against IgY. Methods: three common bacterial pathogens, Staphylococcus aureus (SA), Escherichia coli, (EC), Pseudomonas aeruginosa (PA), were used to prepare the complex antigen. After inactivation, IgY polyclonal antibody was produced by immunizing the egg hens. The polyclonal antibody against IgY was extracted from the immunized eggs. The specificity and concentration titers of IgY antibody were determined by ELISA method. The inhibitory effect of IgY antibody was detected by in vitro inhibition test of SA and EC,PA. Results: SA,EC,PA complex antigen was successfully used to immunize Leghorn laying hens to produce polyclonal antibodies against IgY. ELISA assay showed that the titer of antibody was 1 000 000 and IGY antibody significantly inhibited the growth of the three standard strains of SA,EC,PA. Conclusion: the three bacterial complex antigens of SA,EC,PA can stimulate Leghorn laying hens to produce high titer antibody IgY, which can significantly inhibit the growth of the three standard strains of SA,EC,PA.
