实验感染小鼠肉汁中抗旋毛虫抗体水平的观察

【摘要】:旋毛虫病是一种严重的人兽共患寄生虫病,主要因生食或半生食含有旋毛虫幼虫囊包的猪肉或其它动物肉类而感染。因此,对肉类及肉制品中进行旋毛虫检疫是旋毛虫病防治工作的重要内容。目前用于肉类中旋毛虫检疫的方法有旋毛虫镜检查法和人工消化法,但前者的敏感性低,而后者操作较复杂不易推广应用。由于IgG抗体在宿主血液和组织液内均有分布,应用免疫学方法从动物肉汁中进行抗旋毛虫抗体的检测有望用于旋毛虫的检疫。然而,关于动物感染旋毛虫后肉汁中抗体水平的动态变化尚未见报道。鉴于此,本文应用旋毛虫小鼠动物模型,通过间接ELISA对不同剂量旋毛虫感染小鼠后不同时间血清和肉汁的抗体水平进行了研究;并对小鼠肌肉在不同温度下保存不同时间后肉汁抗体水平的动态变化进行了观察。 材料和方法 1.对ELISA检测旋毛虫感染小鼠肉汁抗体的最适条件进行了选择,包括抗原的最适包被浓度及血清最佳稀释度,酶标二抗、肉汁最佳稀释液及最佳稀释度的选择。 2.将300只健康昆明小鼠随机分成3组,每组100只。每组分别感染旋毛虫100条/只(轻度感染组)、300条/只(中度感染组)和500条/只(重度感染组),观察小鼠感染不同剂量旋毛虫后不同时间(1-18周)血清及肉汁抗体水平的变化。 3.将32只健康昆明小鼠随机分为4组,每组8只。每只分别感染50、100、300、500条旋毛虫,感染6后周观察小鼠膈肌虫荷与血清及肉汁抗体水平的关系。 4.将70只健康昆明小鼠随机分成5组,每组14只。5组分别感染不同种的旋毛虫(旋毛虫、乡土旋毛虫、布氏旋毛虫、伪旋毛虫、纳氏旋毛虫),,每只感染500条幼虫。感染后
[Abstract]:Trichinellosis is a serious zoonotic parasitic disease which is mainly infected by raw or half-eaten pork or other animal meat containing the cyst of Trichinella spiralis larvae. Therefore, the quarantine of Trichinella spiralis in meat and meat products is an important part of trichinellosis control. At present, the quarantine methods of Trichinella spiralis in meat are Trichinella spiralis microscopic examination and artificial digestion, but the former has low sensitivity, and the latter is difficult to be popularized and applied in complicated operation. Because of the distribution of IgG antibody in host blood and tissue fluid, the detection of anti-Trichinella antibody from animal gravy by immunology is expected to be used for quarantine of Trichinella spiralis. However, the dynamic changes of antibody levels in succulent juice of animals infected with Trichinella spiralis have not been reported. In view of this, an animal model of Trichinella spiralis (Trichinella spiralis) was used to study the antibody levels in serum and gravy of mice infected with Trichinella spiralis (Trichinella spiralis) at different time points by indirect ELISA. The dynamic changes of antibody level of succulent were observed after the muscle was stored at different temperatures for different time. Materials and methods 1. The optimum conditions for detection of antibody against Trichinella spiralis infection in mouse gravy by ELISA were selected, including the optimum coating concentration of antigen and the best dilution of serum, enzyme-labeled secondary antibody, The best diluent and the best dilution of gravy. 2. 300 healthy Kunming mice were randomly divided into 3 groups with 100 mice in each group. There were 100 Trichinella spiralis in each group (mild infection group), 300 Trichinella spiralis (moderate infection group) and 500 Trichinella spiralis (severe infection group). The changes of antibody levels in serum and gravy of mice infected with different doses of Trichinella spiralis were observed at different time (1-18 weeks). 3. 32 healthy Kunming mice were randomly divided into 4 groups with 8 mice in each group. Each Trichinella spiralis was infected with 50100300500 Trichinella spiralis respectively. Six weeks after infection, the relationship between phrenic myomyotic charge and antibody levels in serum and gravy in mice was observed. 4. Seventy healthy Kunming mice were randomly divided into 5 groups with 14 mice in each group. Five groups were infected with different species of Trichinella spiralis (Trichinella spiralis, Trichinella bruciformis, Trichinella pseudotinalis, Trichinella nahrensis), each infected with 500 larvae.
